  Aidsmap | Autologous CD8 T-cell infusion
CD8 T-cells are immune cells which play a key role in the cellular immune response (as opposed to the humoral immune response, which involves antibodies).
CD8 T-cells also seem to have non-cytotoxic activity against HIV - that is HIV replication is stopped without infected cells being killed.
Walker (1986) reported that CD8 lymphocytes are capable of suppressing the replication of HIV in autologous peripheral blood mononuclear cells of patients with AIDS.

However, there is also some evidence from animal models of human autoimmune diseases, such as arthritis, that suggests that T cells which recognize mycobacterial hsp60 and may sometimes cross-react with self hsps and cause disease (Holoshitz et al., 1983 and 1984; Shinnick et al., 1988; van Eden et al., 1988; Young et al., 1988).
We have recently demonstrated that hsp70 fusion proteins can elicit CD8+ CTL in the absence of CD4+ T lymphocytes and that this function resides in a 200-amino acid segment of TBhsp70, indicating that chaperone activity is not required (Huang et al., 2000).
The similarity of CD8+ CTL responses to OVA.TBhsp70 in CD4-/- and wild-type mice suggests that hsp70 fusion proteins are relatively potent CD8+ CTL immunogens.

Journal of Translational Medicine | Full text | CD8+ T lymphocyte responses target functionally important regions of ...
In HIV-1 infection, virus-specific CD8 T cell responses are readily detected in peripheral blood and lymph nodes, but HIV-1 replication typically persists in the face of an exuberant CD8 response [1-3].
Thus, although the specific relationship between CD8 T cells and viral control in HIV-1 infection remains unclear, CD8 responses appear to be a critical component of an effective HIV-1-specific immune response [16,17].
Although Integrase peptides were targeted by CD8 T cells in HIV-1 infected subjects at three times the frequency of Protease peptides, comparison of CD8 T cell responses per unit protein length suggests that the relative targeting of the two proteins is similar (Figure 6).

A cytotoxic T lymphocyte inhibits acquired immunodeficiency syndrome virus replication in peripheral blood lymphocytes.
CD8+ (suppressor/cytotoxic) lymphocytes block replication of HIV-1 or the simian immunodeficiency virus of macaques (SIVmac) in PBL of infected individuals.
We now show that these CD8+ lymphocytes undergo clonal expansion in vivo after AIDS virus infection of the individual, suggesting they may be antigen-specific T cells.
Finally, physical contact is required for the CD8+ lymphocyte-mediated inhibition of AIDS virus replication, since this inhibitory function is blocked by anti-LFA-1 and anti-CD8 mAbs.

The CD8 antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediates efficient cell-cell interactions within the immune system.
The CD8 antigen, acting as a coreceptor, and the T-cell receptor on the T lymphocyte recognize antigen displayed by an antigen presenting cell (APC) in the context of class I MHC molecules.
Familial CD8 deficiency is a novel autosomal recessive immunologic defect characterized by absence of CD8+ cells, leading to recurrent bacterial infections.

Hence in association with the CD4^+- CD8^+ switch in cured patients it was also observed a slight, but not significant increase of IFN-gamma production at the end of therapy, as well as a striking significant decrease in the IL-4 production in cured patients.
These results suggest that activation of CD8^+ T cells and production of IFN-gamma may have a beneficial effect in ATL, although we can not reject the possibility that the IFN-gamma detected in the culture supernatants would be produced by other cell types (e.g.: natural killer cells).
In this case the CD8^+ T cell response seems to be involved in the mechanisms of protective immunity since there is evidence that the vaccine is able to induce protection in approximately 50% of cases (CMF Antunes et al.

This should not be surprising as CD8+ cells fight many of the microbes that cause diseases in humans.
There were significant associations (P < 0.05) between survival and CD8+ T-cell counts, clinical AIDS stage, risk group, and antiretroviral drug regimen after baseline, but only the use of protease inhibitors had an independent effect on survival (hazard ratio [HR], 0.096; 95% confidence interval [95% CI], 0.094-0.097).
When the study was restricted to patients never exposed to PIs, the parenteral route of disease transmission, high absolute CD8 + T-cell counts, and low serum levels of tumor necrosis factor alpha were independent predictors of survival in extremely advanced HIV1 disease.

In this section, we wish to make two broad points: (i) the host mounts the type of immune response best suited to eradicating the particular type of microbe which it faces; and (ii) the type of immune response mounted is dictated by the interaction of the infectious agent with the host's antigen presentation pathways.
T cells bearing the appropriate TcR are irrevocably committed to divide, expand, develop their effector functions, and pass into the memory phase; it is important to note that no further antigen contact is required.
T cell responses were detected against all 10 known epitopes, presented by at least 6 HLA alleles; promising though this may appear, the responses were not strong, and their induction required three doses of vaccine.

Studies in TCR transgenic mice that expressed the cognate antigen for the transgenic TCR led to the identification of a population of self-specific CD8 T cells that are dependent on interaction with the cognate self-antigen for their development.
Self-specific CD8 T cells comprise ~10% of the CD8 T cells in normal mice and they possess an activated/memory phenotype and exhibit characteristics of both T cells and NK cells.
Furthermore, self-specific CD8 T cells utilize receptors of both the innate and adaptive immune systems in the destruction of syngeneic tumor cells.

FUNCTIONAL ROLE OF CD8+ LYMPHOCYTES IN SIMIAN IMMUNODEFICIENCY VIRUS INFECTION OF RHESUS MONKEYS
Activated CD8+ lymphocytes were present in increased numbers in the paracortex of lymph nodes of SIV-infected rhesus monkeys with a lymphadenopathy syndrome, and SIV was isolated more readily from CD8+ lymphocyte-depleted peripheral blood lymphocytes (PBLs) of SIV-infected animals than from their unfractionated PBLs.
In fact, CD8+ lymphocytes from SIV-infected rhesus monkeys, but not uninfected rhesus monkeys, blocked SIV replication in vitro in PBL populations.
These findings suggest that the CD8+ cell that exerts this effect is either an activated lymphocyte or a memory lymphocyte.
