In all five studies, the percentage of patients achieving headache response 2 hours after treatment was significantly greater among patients receiving ZOMIG Tablets at all doses (except for the 1 mg dose in the smallest study) compared to those who received placebo.
In the two studies that evaluated the 1 mg dose, there was a statistically significant greater percentage of patients with headache response at 2 hours in the higher dose groups (2.5 and/or 5 mg) compared to the 1 mg dose group.
In controlled clinical trials, single doses of 1, 2.5 and 5 mg of zolmitriptan were effective for the acute treatment of migraines in adults.

The long-term efficacy of rizatriptan tablets 5 and 10 mg was investigated in a total of 1854 patients in optional extension phases of 3 Phase III studies.
Approximately 16 150 attacks were treated with rizatriptan tablets 5 mg and approximately 24 043 with rizatriptan tablets 10 mg over a period of up to 12 months (median number of attacks treated per patient was approximately 17).
Event frequencies are calculated as the number of patients who used rizatriptan 5 mg and 10 mg tablets in Phase II and III studies (n=3716) and reported an event divided by the total number of patients exposed to rizatriptan.

Therapeutic Clinical Trials: The efficacy of zolmitriptan conventional tablets in the acute treatment of migraine attacks was evaluated in 5 randomized, double blind, placebo controlled studies, of which 2 utilized the 1 mg dose, 2 utilized the 2.5 mg dose and 4 utilized the 5 mg dose.
In Study 3, which directly compared the 1, 2.5 and 5 mg doses, there was a statistically significant greater proportion of patients with headache response at 2 and 4 hours in the higher dose groups (2.5 or 5 mg) than in the 1 mg group.
Increases in Blood Pressure: In pharmacodynamic studies, an increase of 1 and 5 mmHg in the systolic and diastolic blood pressure, respectively, was seen in volunteers with 5 mg zolmitriptan.

 5-HT1B Receptors and Aggression / Impulsivity -- Neurotransmitter.net
Serotonin 1B (5HT-1B) receptor mediates aggressive behavior in mice models and was proposed to be involved in the control of aggression and impulsivity in humans.
Little is known to what extent behavioral history influences subsequent behavior in mice, therefore the present study investigated the effects of lever-pressing under a fixed-ratio 5 schedule of reinforcement on subsequent differential-reinforcement-of-low-rate (DRL) 36 s performance in wild type mice compared to the behavior of 5-HT1B receptor knockout mice.
All doses caused a significant down-regulation of the 5HT(1B) receptor density in the hippocampal CA(1) and in the medial globus pallidus and a significant up-regulation of the 5HT(2) receptor density in the nucleus accumbens shell.

We report a study of possible association between the polymorphisms in the 5-HT receptor genes (1A, 1B, 1F, and 2A) and suicidal behavior on a sample of 226 suicide victims and 225 healthy control subjects.
We undertook a detailed analysis of expression mapping of the Gi/o-coupled 5-HT1 (5-HT1A, 1B, 1D and 1F) receptor subtypes in the fetal and early postnatal mouse forebrain.
Using receptor subtype-specific riboprobes and in situ hybridization, we observed that all 5-HT1 receptor subtypes are expressed as early as embryonic day (E) 14.5 in the forebrain, typically in gradients within specific structures.

In The Netherlands, Maxalt, a potent, highly selective 5-HT 1B/1D agonist, will be available in 10 mg and 5 mg strengths in tablets and a convenient new formulation, a rapidly disintegrating tablet that dissolves on the tongue and can be swallowed without liquids.
Maxalt was studied for the acute treatment of migraine attacks in four multi-centre, placebo-controlled trials that included more than 2,000 patients who received Maxalt 10 mg or 5 mg for up to one year.
Results from these studies presented at scientific meetings last year showed that Maxalt 10 mg had a rapid onset of action, relieving pain as early as 30 minutes in some patients and providing consistently high rates of relief within two hours.

The triptan antimigraine drugs are derived from the serotonin molecule.
They act on 5-hydroxytryptamine (HT) 1B/1D receptors on blood vessels, trigeminal neurons, and the trigeminal nucleus caudalis, causing constriction of extracerebral intracranial vessels, abolition of dural extravasation and neurogenic inflammation, and inhibition of trigeminal neuronal discharge.
Relpax has more than a 6-fold higher affinity than sumatriptan at the 5-HT1D receptor and more than a 3-fold greater affinity at the 5-HT1B receptor.
