As shown in Figure 3A and 3B, the phase angles of both 5HT-on and MT-on for either PVG rats or LEW rats are remarkably consistent between individuals of the same strain.
For LEW rats (Figure 3B), the 5HT-on ranged from 130 to 172 min (upper panel), and the MT-on spanned from 159 to 179 min (lower panel).
The strain-specific phase angles of 5HT-on and MT-on for PVG and LEW rats represent the extreme types of 5HT-on and MT-on found in SD rats, and are much earlier than those of the Wistar rats (Figure 3D).

The 5HT1 metabolite opens the K+ receptors and initiates hyperpolerization, allowing ionic channels to open and vesicles holding newly created serotonin to release it through the ion channels and become active during a synapse and bind to the terminal buttons where they are used by the transmitter or naturally recycled.
Some studies show the 5HT molecules are also shown to be active in the lessening of the Serotonin 1B's inhibitory properties on serotonin release, providing more of the neurotransmitter to be accessed.
It must be noted as a controversial issue in the medical field that treatment with chemical agents may also delay self-correction of the underlying chemical imbalance, (perhaps caused by a chronic morphological disorder) and growth in the pathways that are implicated in the inefficiency.

4) Acute changes in cortical 5HT may or may not be related to the long-lasting changes that one sees after administering doses of fenfluramine which both interfere with food consumption in animals and at slightly higher doses cause a prolonged depletion of 5HT in brain.
Studies that have been carried out by various investigators who havelooked at 5HT in brain with doses that are very close to the effective dose for suppressing food intake and they have found losses in 5HT for periods from 2 weeks to 17 months depending upon the species involved.
Furthermore the conclusions derived from this data are based on the assumption that the levels of dex-fen and dex-nor-fen are critical determinants of the long-lasting effects on 5HT, the long-lasting effects agreeably being neurotoxic.

BI and BII are examples of 5HT-evoked facilitation of rested synapses in the controls and in the neurons incubated in 100 µM calpeptin for 15 min, respectively.
When calpain activation is inhibited (#8), the impeding substrates are not cleaved, and as a consequence, the translocation of vesicles is slowed down, leading to an increased rate of homosynaptic depression.
Furthermore, following homosynaptic depression, in the presence of calpeptin, 5HT application (#9) and PKC activation (#10) fail to mobilize vesicles, since vesicle translocation is impeded by the hypothetical uncleaved calpain substrate.

Clinical Trial: Hormonal Causes of Menstrual-Related Mood Disorders
The central serotonergic (5HT) system is one of the candidate systems that may mediate the abnormal mood and behavioral response to ovarian steroids observed in women with menstrual-related mood disorders (MRMD).
Numerous studies have attempted to measure 5HT system function in women with MRMD and controls; however, previous studies were restricted to the evaluation of indirect measures of 5HT function (e.g., platelet 5HT uptake or responses to serotonergic pharmacologic challenges).
Radioligand imaging of specific components of the 5HT system currently represents the most direct measure of central serotonergic function available.
