Adjust the Spec 20 to zero absorbance using cuvette B, filled with the solution from tube 1.
Read off the absorbance value where the linear portion of the line ends and note the time in minutes.
Divide the absorbance value by the time in minutes to obtain absorbance change/min; thisis the reaction rate.

The infrared absorbance spectrum of the sample melted and prepared on a caesium or potassium bromide plate corresponds to the spectrum in the Appendix
The absorbance of the solution of the solvent residue (except for methyl alcohol) shall not exceed 0.01 per cm path length between 280 and 400 nm.
Before proceeding with the analysis of a sample, determine the absorbance in a 5 cm path cell between 250 nm and 400 nm for the reagent blank by carrying out the procedure, without a wax sample, at room temperature, recording the spectra after the extraction stage and after the complete procedure as prescribed.

 Absorbance
When there is a finite amount of protein (or another analyte that absorbs light), the Transmittance will have a value, 0 < T < 1.
Absorbance is a more useful measurement to quantitatively describe protein concentration because absorbance is directly proportional to concentration:
In a chromatographic separation, the elution of proteins from the chromatography column is frequently followed by absorbance at either 214, 254 or 280 nm, where most proteins absorb strongly.

 Spectrophotometry: Absorbance Spectrum
The absorbance spectrum shows how the absorbance of light depends upon the wavelength of the light.
Enter a wavelength between 380 nm and 780 nm (this is the visible range of the spectrum).
Use the intensity of the transmitted light to calculate the absorbance of the sample at that wavelength.

Absorbance of Feulgen-stained Somatic Cell Nuclei
We can therefore conclude that the absorbing material, that is, deoxyribonucleic acid, is not randomly distributed and consequently that published data on the total nuclear deoxyribonucleic acid content, calculated from cytophotometric measurements of fixed tissue assuming a random distribution of the absorbing particles, are erroneous.
The linear relationship between the nuclear absorbance and the inverse nuclear surface area may simply mean that both the absorbance and the geometrical dimensions are dependent variables of a common variable as, for example, the growth process, thus :
A practical consequence of this is, as is also evident from the equation, that a simple measurement of the nuclear geometrical dimensions will yield the same information as that obtained by the far more complicated and costly photometric method.

Daily outdoor, indoor and personal PM and absorbance (proxy for elemental carbon) concentrations were measured among elderly subjects with cardiovascular disease in Amsterdam, the Netherlands, and Helsinki, Finland, during the winter and spring of 1998–1999 within the framework of the ULTRA study.
Traffic was also an important determinant of absorbance: living near a major street increased exposure by 22%, and every hour spent in a motor vehicle by 13% in Amsterdam.
Cooking was associated with increased levels of both absorbance and PM Our results demonstrate that by using questionnaires in connection with outdoor measurements, exposure estimation of PM and its combustion originating fraction can be improved among elderly persons with compromised health.

Optical time-of-flight and absorbance imaging of biologic media -- Benaron and Stevenson 259 (5100): 1463 -- Science
Imaging the interior of living bodies with light may assist in the diagnosis and treatment of a number of clinical problems, which include the early detection of tumors and hypoxic cerebral injury.
An existing picosecond time-of-flight and absorbance (TOFA) optical system has been used to image a model biologic system and a rat.
Model measurements confirmed TOFA principles in systems with a high degree of photon scattering; rat images, which were constructed from the variable time delays experienced by a fixed fraction of early-arriving transmitted photons, revealed identifiable internal structure.

if you use dilution factor 50 instead of 100, you should have twice the concentration, which will mean twice the absorbance.
Dilute your DNA less so that the OD readings are in the range of 0.1 - 1.0.
Therefore you should dilute less the DNA and remeasure it but you shouln´d expect that 2 x more concentrated DNA will give you 2 x the absorbance.
