This problem is most pronounced at low wavelength, since the pi to pi* absorption band of the peptide bond can be quite sharp and have a steep shoulder.
My recommendation is to make a wavelength scan of the sample and find the exact peak of the absorption.
You can find out the effect for yourself by finding the point of the steepest slope in the absorption band and measure a scan across the entire cell by running it at a slow speed so it doesn't sediment.

Hi, as a newcomer in XL-A technique I have to learn something what I have not observed in such extent with the Model E using absorption or Rayleigh inter- ference optics.
For the experiments of heterologous association (different proteins or protein and nucleic acid) we need exact information about the molecular mass of the reactants under the choosen conditions and also the initial concentrations.
From these observations I would like to ask the XL-A user for similar experience or to know until to which absorption (0.8, 1.0 or somewhat higher) the Lambert law is valid for the concentration determination in XL-A? Best regards.

The nonlinear optical absorption, scattering and optical limiting properties of CdS nanoparticles dispersed in dimethylformamide (DMF) are investigated.
The nanoparticles are synthesized using the standard chemical synthesis method with thioglycerol as the capping agent.
Strong two-photon absorption and nonlinear scattering are found to be responsible for good optical limiting characteristics in these nanoparticles.
