In proteomics, an activator is a type of effector that increases the rate of enzyme mediated reactions.
In genetics an activator is a DNA-binding protein that regulates one or more genes by increasing the rate of transcription.

ERK and JNK signaling pathways are involved in the regulation of activator protein 1 and cell death elicited by three isothiocyanates in human prostate cancer PC-3 cells.
The activator protein 1 (AP-1) and MAPK signaling pathways are believed to play an important role in cancer chemoprevention and chemotherapy due to their involvement in tumor cell growth, proliferation, apoptosis, and survival.
Taken together our results indicate that the activation of the ERK and JNK signaling pathways is important for transcriptional activity of AP-1, and is involved in the regulation of cell death elicited by ITCs in PC-3 cells.

Expression and specificity of human GM2 activator protein.   (Site not responding. Last check: 2007-10-31)
The effectiveness of GM2 activator in stimulating the hydrolysis of GM2 may be due to its ability to recognize the specific trisaccharide structure of the GM2 epitope, GalNAc beta 1-->4(NeuAc alpha 2-->3)Gal-, and to modify the GalNAc-NeuAc interaction in this structure.
Taken together, our results suggest that the mechanism of action of GM2 activator is different from saposin B, and the action of GM2 activator is more than to solubilize lipid substrates.
The recombinant GM2 activator was found to be as active as that isolated from human kidney.

The GM2 activator protein, a novel inhibitor of platelet-activating factor.
The GM2 activator protein is required as a substrate-specific cofactor for beta-hexosaminidase A to hydrolyze GM2 ganglioside.
The inherent stability and protease resistance of the small, monomeric GM2 activator protein, coupled with the ability to produce large quantities of the functional protein in transformed bacteria, suggest it may serve as such an agent.

Binding between co-activator protein and the ER: 17b -estradiol complex is also an essential step in a process leading to cell division.
We have used computer models of binding between the ER and co-activator protein to design organic molecules that may bind to the ER at the same position as the co-activator protein.
We are attempting to design and identify chemicals that bind to a different part of the ER and which like tamoxifen block formation of the ER: co activator protein complex.

In the absence of the inducer, quinic acid, the repressor protein binds to the activator protein, blocking transcription of the qa genes.
Addition of quinic acid releases the activator protein, which is then free to bind to its activator binding sites, increasing the levels of transcription of the qa genes.
The activator binding sites are composed of a 16 base pair (bp) conserved sequence, but the importance of the individual bases in the sites is currently unknown.

The Heme Activator Protein Hap1 Acts as a Transcriptional Repressor in Saccharomyces cerevisiae.
The heme activator protein Hap1 is known to be a potent transcriptional activator of many genes encoding functions required for respiration and for controlling oxidative damage.
As an activator, the activity of Hap1 is stimulated by heme and is highly sensitive to changes of heme concentration.

Transcription activation is a complex process brought about by a combination of different proteins.
Transcription activation by the catabolite activator protein (CAP; also known as cAMP receptor protein, CRP) is one of the simplest gene activation models in bacteria, requiring only CAP and RNAP protein factors for transcription at CAP-dependent promoters; by comparison, transcriptional activation in eukaryotes can require dozens of proteins involving several DNA sites.
For instance, in the presence of glucose, the intracellular level of cAMP drops, repressing transcription at the lac operon; in the absence of glucose, intracellular cAMP levels rise, and the lac operon is activated in response to cAMP-induced CAP binding.

Luciferase assays demonstrated that SRAP enhances the transactivation activity of the AR, the glucocorticoid receptor and the peroxisome proliferator-activated receptor gamma(1) in a ligand-dependent manner.
Using a green fluorescent protein (GFP) fusion-protein construct, we demonstrated in vivo translation of the GFP-SRAP fusion protein in HeLa cells co-transfected with pSG5AR and reporter gene in the presence of 5 alpha-dihydrotestosterone (DHT).
Co-transfection of the GFP-SRAP fusion protein expression plasmid enhanced the transactivation activity of AR whereas incorporation of mutations in SRAP of the fusion protein resulted in loss of enhancement of the transactivation activity.

Supershift assays revealed that fra-1 participates in the activator protein 1 complex together with c-fos, c-jun and junB.
Transient transfection experiments using a reporter plasmid containing an activator protein site upstream of the CAT reporter gene revealed that the phorbolester-induced CAT activity was suppressed by retinoids.
Since fra-1 lacks a transactivation function, we therefore suggest that the retinoid-mediated induction of fra-1 might function as a negative regulator of the activator protein 1 activity in human pancreatic carcinoma cells.

Because of its role in delivering messages from the primary glucose sensor (adenyl cyclase) to the synthetic machinery, cyclic AMP is often known as a second messenger.
Catabolite activator protein (CAP), also known as cyclic AMP receptor protein (CRP), is activated by cyclic AMP and stimulates synthesis of the enzymes that break down non-glucose food molecules.
CAP binds to a specific DNA sequence, which is found next to the genes that are activated.

To determine the effects of oxidized LDL on macrophage gene expression, macrophages incubated with native or oxidized LDL were analyzed by differential display technique.
Quantitative RT-PCR data using specific TaqMan probe showed the increased GM2 activator protein mRNA expression in macrophages incubated with oxidized LDL by about twice (215.4˜31.6 %) of that in macrophages incubated with native LDL.
Our results shows that ox-LDL upregulates GM2 activator protein gene expression, indicating that GM2 activator protein may be associated with the pathogenesis of atherosclerosis.
