Myofibril - Wikipedia, the free encyclopedia
Thick filaments consist primarily of the protein myosin, held in place by titin filaments.
The protein complex composed of actin and myosin is sometimes referred to as "actomyosin."
In striated muscle, such as skeletal and cardiac muscle, the actin and myosin filaments each have a specific and constant length on the order of a few micrometers, far less than the length of the elongated muscle cell (a few millimeters in the case of human skeletal muscle cells).

This transformation occurs in the actomyosin complex, aligned into the filaments in the heart, as a result of cyclic interaction of myosin heads with actin filaments [Huxley, 1990,Irving, 1985,Saeki, 1995].
This hierarchical structure incorporates the cross-bridge state distribution functions and the variables describing the activation of the actomyosin complex by calcium ions as used in [Vendelin et al., 2000].

While on the microscale the axon clearly functions as a kind of wire that propagates an electric current to the synapse, on the nanoscale it serves the purpose of transporting vesicles containing the neurotransmitters required for signal transduction across the synapse.
The vesicles (nanoscale vessels or containers) are transported by the actomyosin molecular motors described on the previous page.
Actin and myosin are the two components of actomyosin, which is a ubiquitous complex used for muscle contraction in addition to its vesicle transport function in neurons.

The actomyosin system (Figure 1) has been studied extensively as a model of chemomechanical transduction, with a major focus on the conformational changes arising within actin and myosin that lead to force generation.
My work focuses on: (1) Determining residues in actin that are important in the actomyosin interaction; and, (2) Testing the hypothesis that specific charged residues in the N-terminus of actin are important in the transition from weakly- to strongly-bound actomyosin complexes.
Since the transition from weakly- to strongly-bound actomyosin states directly precedes the powerstroke, free energy resulting from this transition (A-M-ADP-Pi to A-M*-ADP-Pi) is considered to be instrumental in the transduction of the chemical energy of ATP to mechanical force.

Hasselbach (1956) reported an excellent correlation between the rate of NTP hydrolysis by actomyosin and the isometric tension of glycerinated muscle fibers.
After the success of fluorescence microscopy in visualizing fluorescently labeled single actin filaments (Yanagida et al., 1984), a more versatile and stable assay was developed wherein actin filaments move over surfaces coated with myosin filaments (Kron and Spudich, 1986), or proteolytic fragments of myosin (Toyoshima et al., 1987).
First, it has been shown that the velocity of actin filaments sliding over myosin-coated surfaces and the velocity of myosin-coated beads along actin cables is analogous (with slight differences, depending on conditions) to the speed of unloaded shortening of muscle fibers (Sheetz et al., 1984; Homsher et al., 1992).

Research and Reviews: Meat 2001, Special Circular 183-02
The physicochemical state of the myofibrillar proteins, myosin and actomyosin, confer functionality to meat systems and thereby have a direct role in determining the quality and value of processed meat (Yasui et al., 1982; Camou and Sebranek, 1991).
Thus, myosin and actomyosin are the most important factors for the development of the binding properties of thermally induced meat protein gels (Samejima et al., 1969; Samejima, et al., 1981 and 1982).
Not only do myosin and actomyosin contribute to the rigidity (firmness) of thermally induced meat gels, they are also critical to the water holding capacity (WHC) and high cooking yields of comminuted meat products such as sausages (Hamann, 1988; Smith, 1988).

Phosphorylation of myosin light chains in mouse fast-twitch muscle associated with reduced actomyosin turnover rate -- ...
Phosphorylation of the 18,000-dalton light chains of the fast-twitch myosin in mouse extensor digitorum longus muscles was correlated with reduction in the rate of the actomyosin adenosinetriphosphatase in vivo, but neither of these changes occurred in the soleus muscle.
These results suggest that actomyosin interactions can be down-regulated by a reversible covalent modification of myosin light chains, that a mechanism for thick-filament regulation occurs in vertebrate skeletal muscle, and that the expression of this regulation may be limited to a specific fiber type.

While actomyosin regulation and microtubule bundling may be the primary regulatory roles of these factors, there are additional ways that the Pbl RhoGEF and RacGAP50C could influence both the actin and microtubule-based cytoskeleton.
Although the molecular mechanism behind this reorganization of the actomyosin cytoskeleton is not known, it indicates that the motor protein is able to mediate associations between the actomyosin and microtubule cytoskeletons.
The formation and maintenance of both the central spindle and the actomyosin ring could be mediated by physical interactions between interzonal microtubules and components of the contractile ring.
