Tumour suppressors: Adaptor protein connects to cancer - Cell Signaling Update - Signaling Gateway
Alexandra Flemming and colleagues propose that an adaptor protein that is expressed during B-cell development — the SLP65 protein — is a tumor suppressor that limits the proliferation of precursor B cells and promotes their differentiation into mature cells.
Adaptor proteins mostly function as flexible molecular scaffolds that mediate protein–protein and protein–lipid interactions in signalling pathways.
Writing in Nature Immunology, Alexandra Flemming and colleagues propose that an adaptor protein that is expressed during B-cell development — the SLP65 protein — is a tumour suppressor that limits the proliferation of precursor B cells and promotes their differentiation into mature cells.

Adaptor proteins which may be components of the PTK/adaptor protein complexes of the invention, may include, for example, SHC, ISGF3.alpha., and members of the GRB subfamily of proteins, such as GRB-1/p85, GRB-2, GRB-3/c-Crk, GRB-4/Nck, GRB-7, and GRB-10, with GRB-7 being a preferred adaptor protein component of the PTK/adaptor protein complex of the invention.
New members of the PTK and/or adaptor protein families capable of forming the complexes of the invention may be identified and isolated by molecular biological techniques well known in the art.
Briefly, new members of the adaptor family of proteins may be identified by their ability to specifically bind to at least a portion of a tyrosine-phosphorylated peptide comprising an adaptor-protein-binding region.

Energy Citations Database (ECD) - Energy and Energy-Related Bibliographic Citations
Adaptor proteins are molecules with multiple protein interaction motifs that do not appear to have any catalytic activity of their own but mediate the interaction of other proteins.
While the exact function of the SHB proteins has not been determined yet, it is likely that they play an important role in the transduction of signals from a tyrosine kinase receptor to an unknown cytoplasmic enzyme.
As the proteins of signal transduction pathways can be involved in human pathology, particularly oncogenesis, it is important to map their genes to determine proximity to known sites of mutation, such as loss of heterozygosity or chromosomal translocation.

Negative regulation of PI 3-kinase by Ruk, a novel adaptor protein
protein corresponded to a molecular mass of 85 kDa, which is much higher than that predicted from the amino acid sequence, suggesting the existence of post-translational modifications or a conformation that retards mobility.
In addition to these structural modifications, interactions between PI 3-kinase subunits and membrane-associated receptors or adaptor proteins have another important function in vivo, namely translocation of the enzyme to the membrane where the lipid substrates are localized (reviewed in Wymann and Pirola, 1998; Leevers et al., 1999; Rameh and Cantley, 1999).
Proteins were also immunoprecipitated from cell lysates using specific antibodies, and protein G–Sepharose was used to bring down the immune complexes.

Oxidized proteins appear to be relatively poor substrates for ubiquitination, and the ubiquitination system does not seem to be involved in the recognition or targeting of oxidized proteins.
A search for such proteins was initiated because it became impossible to reconcile a bewildering spectrum of experimental observations relating to various functions of LRP and other members of the LDL receptor gene family with a simple role as an endocytic receptor or cellular transporter of extracellular ligands.
These newly identified adapter and scaffold proteins are key organizers or regulators of MPAK signaling, ion channel function in the postsynaptic density, microtubular transport, nitric oxide signaling, and axon guidance.

We screened a cDNA expression library with the EH domains of intersectin, and identified two the mouse homologue of rat Epsin, an Eps15-binding protein recently implicated in clathrin-mediated endocytosis (Chen et al., Nature, 1998).
