JGV Direct - Review: Adenovirus genomes (A. J. Davison and others)
Capture and duplication of genes in the setting of a 'leader–exon structure', which results from widespread use of splicing, appear to have been central to adenovirus evolution.
Members of the family Adenoviridae are non-enveloped, icosahedral viruses that replicate in the nucleus.
Their linear, double-stranded DNA molecules are 26–45 kbp in size and rank as medium-sized among the DNA viruses.

It is shown here that E1a has the two functions characteristic of a typical cellular activator: one direct E1a to the promoter, perhaps by interacting with a DNA-bound protein, and the other, an activating region, enables the bound activator to stimulate transcription.

Technical Abstract: The Adenoviridae family was originally divided into two genera, Mastadenovirus and Aviadenovirus.
Division was further supported after genomic analysis demonstrated marked differences in the two subgroups.
A new genus, Atadenovirus, within the family Adenoviridae has now been created to accommodate this unique adenovirus cluster.

 Family Adenoviridae
The Family Adenoviridae is a large family of non-enveloped, icosahedral viruses with linear, double stranded DNA genomes.
This, taken together with their great diversity and relative ease of manipulation, have made adenoviruses an attractive tool for gene therapy and cancer treatments.
Human adenovirus type 2 (Ad2) is the best studied strain of adenoviruses and is considered the prototype virus for the family Adenoviridae, so the information presented will be for the Ad2 virus unless it is stated otherwise.

In the passed decade, the Adenovirus Study Group has characterized the genome of new animal adenovirus isolates and compared them to the well-known mammalian adenoviruses.
Based on the DNA sequences, the recognition of a unique genomic organization and the results of phylogenetic analyses, adenoviruses have been divided into separate taxons equivalent to the existing two (Mastadenovirus & Aviadenovirus) genera within the family Adenoviridae.
