  MAPS - Volume 6 Number 2 Winter 1995-96 - 4-methyl aminorex   (Site not responding. Last check: 2007-10-13)
4-methyl aminorex produces a very long-lasting experience of about sixteen hours that is characterized by an increase in attention and an enhanced ability to recall, analyze and process information.
What distinguishes 4-methyl aminorex from other substances like caffeine and nicotine, which also have these effects though to a lesser degree, is its effect on emotions.
4-methyl aminorex may be the single most important drug for which research is not currently being attempted.

Aminorex, also called aminoxaphen, 2-amino-5-phenyl-2-oxaoline, or 4,5-dihydro-5-phenyl-2-oxazolamine, is a phenethylamine in which the side chain has been cyclized into a substituted oxazoline.
Aminorex is chemically and pharmacologically similar to amphetamine, methamphetamine, and cis-4-methylaminorex, all of which are controlled substances with high abuse potential.
Any person who manufactures, distributes, delivers, imports or exports aminorex or who engages in research or conducts instructional activities with respect to aminorex, or who proposes to engage in such activities, must be registered to conduct such activity in accordance with parts 1301 and 1311 of title 21 of the Code of Federal Regulations.

A 4-methyl derivative of aminorex has recently appeared on the clandestine market as a designer drug.
In the present study, the stimulus effects of aminorex itself were evaluated in rats trained to discriminate either 0.75 mg/kg S(+)-amphetamine or 1.5 mg/kg fenfluramine from saline.
Thus, the stimulus effects of aminorex are qualitatively similar to those of amphetamine and unlike those of fenfluramine.

Dexfenfluramine   (Site not responding. Last check: 2007-10-13)
In the late 1960's, the widespread use of an appetite suppressant, aminorex, caused an epidemy of primary pulmonary hypertension, a deadly disease with no effective treatment available.
Using the whole-cell patch clamp technique, we found that anorectics inhibit potassium channels in the cell membrane of the smooth muscle cells of resistance pulmonary arteries, causing membrane depolarization and vasoconstriction (Figure 1).
This is a result of a patch-clamp experiment where the composition of the patch pipette solution and the bathing solution ensured that the current was carried exclusively by potassium.

Over the next six years, an Aminorex related epidemic of primary pulomnary hypertension raged in Europe where a ten-fold increase in primary pulmonary hypertension cases is documented.
Half of the PPH patients died within ten years and the rest suffered significant oxygen deprivation and are debilitated for the remainder of their lives.
Pondimin is very similar to Aminorex and is in the same chemical family.

Between 1967 and 1973, an unexplained increase in the incidence of Primary Pulmonary Hypertension (PPH) was reported in central Europe.
This increase was eventually attributed to aminorex fumarate, an amphetamine-like drug introduced in Europe in 1965 to control appetite.
When aminorex was removed from the market, the incidence of PPH went down to normal levels.

A case in which aminorex (cis ± 4,5-dihydro-5-phenyl-2-oxazolamine) was prepared as a “designer” analog of a controlled substance, methylaminorex (cis ± 4,5-dihydro-4-methyl-5-phenyl-2-oxazolamine) is described.
The confiscated drug sample was analyzed by high-field (300-mHz) proton nuclear magnetic resonance (NMR) and carbon-13 NMR spectroscopy, as well as electron impact mass spectrometry (70 eV).
These examinations proved conclusively that the material in question was aminorex.
