  NationMaster - Encyclopedia: Azapirone
Azapirones are a class of drugs with anxiolytic effects used in the treatment of anxiety.
Azapirones are agonists of serotonergic neurotransmission at the 5-HT 1A receptor.
The azapirone compounds of Formula I in the context of the '325 application include buspirone, and claim 7 of the '325 application is specifically directed to buspirone.

explicitly stated on the record that Schwimmer was the sole inventor of the use of most of the azapirone compounds covered by the count except for buspirone in the treatment of sleep apnea.
There are two aspects to the use of azapirones in treating sleep apneas.

The pharmaceutical composition defined in any one of claims 1-4 wherein the unit dosage form containing the nicotine, at least one active nicotine metabolite, a combination of nicotine and an active nicotine metabolite, or an azapirone, or a pharmaceutically acceptable salt thereof is contained in a patch for transdermal delivery.
The pharmaceutical composition defined in any one of claims 2-4 or 7 wherein the nicotine, at least one active nicotine metabolite, a combination of nicotine and an active nicotine metabolite, or an azapirone, or a pharmaceutically acceptable salt thereof is included within the reservoir containing the bupropion base in the transdermal patch.
There is no mention of administration of the azapirones by any route in conjunction with transdermal administration of bupropion base.

Azapirones (buspirone etc) for generalized anxiety disorder (GAD)
Whether azapirones are useful as first line treatment in general anxiety disorders still needs to be answered.
Azapirones, including buspirone, were superior to placebo in treating GAD.
The calculated number needed to treat for azapirones using the Clinical Global Impression scale was 4.4 (95% confidence interval (CI) 2.16 to 15.4).

IngentaConnect Prediction of the Affinity of the Newly Synthesised Azapirone Der...   (Site not responding. Last check: )
The performance of artificial neural network (ANN) in predicting the affinity of a series of 65 new azapirone derivatives for rat brain serotonin 5-HT1A receptors based on high-performance liquid chromatography (HPLC) retention data and on non-empirical structural parameters of the compounds' was studied.
However, the percent of correctly classified compounds was higher in the case of the ANN processing of the non-empirical structural descriptors of azapirone derivatives.
Since the ANN analysis of the retention data and of the structural parameters originating from calculation chemistry allows to segregate drug candidates according to their pharmacological properties that, in consequence, may be of help to limit the number of biological assays in the search for new drugs.

 Azapirones for generalized anxiety disorder
From the results of 36 randomized controlled trials, azapirones appear to be superior to placebo in short-term studies (four to nine weeks) but may not be superior to benzodiazepines.
Azapirones are a group of drugs that work at the 5-HT1A receptor and are used to treat patients suffering from generalized anxiety disorder (GAD).
Azapirones appeared to be useful in the treatment of GAD, particularly for those participants who had not been on a benzodiazepine.

Serotonin re-uptake inhibitors like fluoxetine induce a serotonergic akathisia which can be blocked by buspirone.
When SSRI compounds stop working with re-exacerbation of depression, adjunctive use of azapirones may theoretically extend their action.

Make sure you discuss these risks with your health care professional, and if you are taking benzodiazepines, talk to your health care provider before you stop taking them.
Azapirone is a class of drugs that includes buspirone (BuSpar), approved for generalized anxiety disorder, because they enhance the activity of serotonin.
It usually takes several days to weeks for the drug to be fully effective, and it is not useful against panic attacks.
