Subsequent to August 1929, Barker was permanently on the Asiatic Station and operated with the destroyer divisions of the Asiatic Fleet.
On 7 December 1941 Barker was at Tarakan, Borneo, and upon receipt of the news of the Pearl Harbor attack, immediately commenced patrolling the surrounding area.
Barker was badly shaken by near misses during the 15 February action and required emergency repairs.

 USS Alden (DD-211) - Encyclopedia, History, Geography and Biography
Alden accompanied the squadron flagship Paul Jones (DD-230), her division-mates Whipple and Barker (DD-213), to sea from their base at Chefoo, and rendezvoused with Admiral Yarnell's flagship, Augusta (CA-31), at the end of the afternoon watch on 25 July.
Early on the morning of 11 December 1937, Alden and Barker, then at Manila, received orders to proceed immediately to the aid of the American Dollar liner SS President Hoover, which had run aground off Formosa.
Barker arrived soon thereafter, after which time an officer from Ashigara arrived on board Alden to give his government's permission to enter and assist President Hoover.

Warfare between local warlords around Shanghai in late 1924 and early 1925 resulted in Whipple's being called upon to serve as a transport.
The three destroyers landed the marines on 22 January, relieving the 28-man detachment from the gunboat at that time.
The destroyer—in company with sisters Alden (DD-211), Barker (DD-213), and Paul Jones (DD-230)—rendezvoused with Fleet flagship Augbsta (CA-31), on the 25th, en route to the coast of Siberia.
