Basophils are similar in appearance to eosinophils except that their cytoplasm contains blue or purple staining granules.
Basophils have relatively large, sparse, cytoplasmic granules which stain dark purple with basic (alkaline) dyes, and tend to disappear in films stained with new methylene blue.
The characteristic metachromasia of basophils and mast cells is ascribed to their granule content of sulfated glycosaminoglycans (mucopolysaccharides), particularly heparin or chondroitin sulfate and dermatan sulfate, depending on the species.
