 Beta Galactosidase
Beta galactosidase the molecule rotating on the left is an enzyme that initiates the breakdown of the sugar lactose.
Beta galactosidase also forms a complex with ONPG molecules and is able to hydrolyze them.
This bridge can be broken when the enzyme beta galactosidase binds to lactose and a water molecule reacts with the oxygen atom in the bridge.

 lacZ -
Germ line transformation experiments with the upstream region of the D.melanogaster beta 2 tubulin gene fused to the Escherichia coli lacZ gene resulted in the correct tissue specific expression of the reporter gene.
The lethal E phi lacZ fusions are also lethal to a mutant, designated slyD, which was isolated as a spontaneous E. coli mutant resistant to the expression of the intact E gene.
The strains of E. coli that we have constructed are temperature-sensitive for the Mcr and Mrr systems and have been further modified to include a lacZ gene fused to the damage-inducible dinD locus of E. coli.

 Lactase enzyme supplement (Beta-galactosidase)
Deficiency of intestinal beta galactosidase activity causing intolerance to milk-containing foods which presents clinically as abdominal cramps, diarrhoea and bloating.
Diagnosis today is made by a positive lactose H2 breath test but if contrast studies are done, the addition of lactose to the barium produces barium dilution, bowel dilatation and rapid transit.
Generally, however, lactase usage has not reached its full potential, as present enzymes are relatively expensive and can only be used at low temperatures.

In contrast, Suffolk Red grapes contained higher beta-galactosidase activity (186 units per mg protein), but had a much lower firmness value (less than 1 N).

The results of this study indicate that there is an inverse relationship between the activity of beta-galactosidase and the firmness of mature grapes.

BTP - Current Students - Emilie Porter
Active beta-peptides induce the leakage of beta-galactosidase from peptide-treated B. subtilis cells, as do alpha-helical antibiotic peptides, and this similarity suggests that the beta-peptide mode of action involves disruption of microbial membranes.
We have previously identified a 17-residue beta-peptide (called beta-17) with antibiotic activity similar to that of a magainin derivative against a variety of bacteria, including two clinical isolates that are resistant to common antibiotics.
We have designed beta-peptides that are helical, cationic and amphiphilic with the intention of mimicking the biological activity of amphiphilic, cationic alpha-helical antimicrobial peptides found in nature (e.g., magainins).

In whole animal cell sorting (WACS), live cells derived from animals harboring a lacZ transgene are purified according to their level of beta-galactosidase expression with a fluorogenic beta-galactosidase substrate and fluorescence-activated cell sorting.
Detection for inversional activation of this gene is achieved by a fluorogenic assay, termed FACS-Gal, that detects beta-galactosidase activity in viable cells.
In the second vector, the activatable gene is the bacterial beta-galactosidase gene (lacZ).
