Presence of extended spectrum betalactamases (ESBL) was inferred by both double disk diffusion tests and minimum inhibitory concentration (MIC) of third and fourth generation cephalosporins alone and in the presence of clavulanic acid.
CONCLUSION: The broad extended spectrum betalactamase within genus Proteus was TEM-1, while CTX-M-2 was the ESBL responsible for the third and fourth generation cephalosporins in P. mirabilis.
In P. vulgaris and P. penneri this resistance was associated with the hyperproduction of the chromosomal encoded betalactamase.

Only 17.5% of betalactamase producers could be detected by one of the three methods used whereas 75.0% positivity was obtained by using all three methods.
Totally, 18.4% of the strains were sensitive to penicillin by disc diffusion but were positive for betalactamases indicating insufficient amount of the enzymes to hydrolyse penicillin.
In contrast, 22.6% were positive for betalactamase production even though they were sensitive to penicillin by disc diffusion technique.

Using this approach, we were able to introduce beta-lactamase activity into the metallohydrolase scaffold of glyoxalase II.
The resulting enzyme, evMBL8 (evolved metallo betalactamase 8), completely lost its original activity and, instead, catalyzed the hydrolysis of cefotaxime with a (kcat /Km)app of 1.8 x 102 (mole/liter)-1 second-1, thus increasing resistance to Escherichia coli growth on cefotaxime by a factor of about 100.
