  036: CLONING AND CHARACTERIZATION OF A BETARETROVIRUS INFECTION IN PATIENTS WITH PRIMARY BILIARY CIRRHOSIS   (Site not responding. Last check: 2007-11-06)
Serologic studies showed that 42 of 59 PBC patients had Western blot reactivity to betaretrovirus proteins as compared to 3 of 23 liver disease controls and 2 of 16 blood donors (p < 0.001).
Immune purified anti-mitochondrial antibodies from PBC patients' sera were found to bind to pure virus Western blots, which was abrogated by pre-incubation of serum with the mitochondrial autoantigens.
Serologic and tissue evidence for viral infection was found in ~75% of patients with PBC and was frequently observed in control subjects as well.

In my research, I have recently identified a human betaretrovirus associated with Primary Biliary Cirrhosis.
Current areas of research include investigating the prevalence of this human betaretrovirus in patients with primary biliary cirrhosis and breast cancer by assessing reverse transcriptase activity, measuring viral levels by real time RT-PCR and standard serologic assays using ELISA and Western blot.
Virus isolation studies are planned to determine Koch’s postulates in vitro using a cell culture model of primary biliary cirrhosis.

 FEBS Abstract
Ribosomal frame-shifts during expression of retroviral proteins provide a unique possibility for covalent joining of nucleocapsid (NC) and dUTPase within Gag-Pro polyproteins.
By developing an antibody against the prototype betaretrovirus Mason-Pfizer monkey virus dUTPase, we demonstrate that (i) the NC-dUTPase fusion protein exists both within the virions and infected cells providing the only form of dUTPase, and (ii) the retroviral protease does not cleave NC-dUTPase either in the virion or in vitro.
We show that recombinant betaretroviral NC-dUTPase and dUTPase have low catalytic activity compared with all other dUTPase.
