  Metabolic Biotinylation
Given metabolic biotinylation allows one to tag a number of capsomer proteins with BAPs and then combine these with any biotinylated ligand, this provides a unique platform to compare the functions of a given viruses proteins for targeting and purification.
As a first example of this approach, we have utilized metabolic biotinylation to directly compare targeted transduction of adenovirus as mediated by its fiber, protein IX, and hexon capsomeres using a variety of biotinylated ligands (Virology 349(2):453-62 2006).
As such, biotinylated vectors can be used as a platform for combinatorial screening of both cell-targeting ligands and for receptors that are compatible with cell targeting.

ARS | Publication request: Lysine-8 and Lysine-12 Are Biotinylated on Human Histone H4.
This study elucidates the lysine residues of Human histone H4 that are the targets for biotinylation and has utilized synthetic peptides to delineate regions of the H4 protein that are biotinylated.
Substitution of lysine-8 or lysine-12 by alanine or arginine decreased biotinylation, suggesting that these lysines are targets for biotinylation; lysine-8 and lysine-12 are also known targets for acetylation.
Here we sought to identify amino acid residues that are biotinylated in histone H4, and we sought to determine whether acetylation and methylation of histones affect biotinylation.

Determination of the Extent of Protein Biotinylation by Fluorescence Binding Assay
The extent of biotinylation was determined by complete acid hydrolysis or by enzymatic digestion using proteinase K to release biotin from the biotinylated proteins, followed by sensitive detection of biotin using a coupled HPLC-binding assay system.
The extent of biotinylation determined by this method was compared with the values obtained by a conventional colorimetric method that is based on the displacement of the dye 4-hydroxyazobenzene-2-carboxylic acid (HABA) from the binding sites of avidin.
It was found that, because the described method determines the amount of liberated biotin after hydrolysis, it does not suffer from steric hindrance problems associated with the ability of biotin on a protein surface to displace HABA from avidin.

In Vitro Enzymatic Biotinylation of Recombinant Fab Fragments through a Peptide Acceptor Tail   (Site not responding. Last check: )
We describe the site-specific enzymatic biotinylation of recombinant anti-estradiol Fab fragments through a 13 amino acid acceptor peptide translationally fused to the C-terminus of the Fd chain.
After the removal of nonbiotinylated Fab fragments by monomeric avidin chromatography, the overall yield of biotinylated Fab was 40%.
The site-specifically biotinylated Fab fragments (BioFab) were tested in streptavidin-coated microtitration wells, to which they were shown to bind linearly with respect to the amount of BioFab added, specifically as indicated by biotin inhibition, and tightly with a half-life of several days.

 Avidity - Biotin AviTag Applications
Because the biotinylation is performed enzymatically, the reaction conditions are very gentle and the labeling is highly specific.
In addition, it should be noted that the Biotin AviTag has only one fifth of the bulk of alternative biotinylation tag sequences that are over 85 amino acid residues long; an important consideration if steric problems are to be minimized.
Fast and efficient in vitro biotinylation is achieved using Avidity's highly purified Biotin Ligase that comes complete with our exclusive reaction mix.
