bumetanide - High Blood Pressure (hypertension) and related information on MedicineNet.com
One mg of bumetanide is approximately equivalent to 10-20 mg of torsemide and 40 mg of furosemide.
Bumetanide was approved for use by the FDA in 1983.
PRESCRIBED FOR: Bumetanide is used in the treatment of mild to moderate hypertension (high blood pressure), and in the management of edema (excessive fluid accumulation) associated with congestive heart failure, renal disease and liver cirrhosis.

 OhioLINK ETD: TILLOTSON, JOHN
Extended-release tablets of bumetanide were developed and evaluated for the impact on bumetanide release by: extended-release polymer concentration, filler excipient type and concentration, pH of dissolution media, ICH accelerated storage conditions, formula reproduction, and media pH and solubility modifiers.
Bumetanide release was slowed as the fraction of the extended-release polymer was increased from 20-60%.
Bumetanide tablets exhibited pH-dependent drug release based upon bumetanide solubility in the study media, releasing slowest in pH 1.5 HCl buffer followed by pH 4.5 phthalate buffer, USP purified water pH 5.8, and pH 6.8 phosphate buffer.

NKCC1 inhibitor bumetanide shows promise for treatment : Epilepsy.com
In a study conducted by Dzhala and coauthors, the "NKCC1 blocker bumetanide shifted ECl negative in immature neurons, suppressed epileptiform activity in hippocampal slices in vitro, and attenuated electrographic seizures in neonatal rats in vivo."
"Bumetanide had no effect in the presence of the GABAA-R antagonist bicuculline, nor in brain slices from NKCC1-knockout mice," test results indicated.
These findings "provide evidence that NKCC1 facilitates seizures in the developing brain, and indicate that bumetanide should be useful in the treatment of neonatal seizures," the researchers concluded.

An existing diuretic may suppress seizures in newborns
The study findings could lead to clinical trials of bumetanide in newborns, whose immature, rapidly-developing brains are especially vulnerable to seizures.
The researchers knew that the diuretic bumetanide inhibits NKCC1 activity in the kidney, and reasoned that the drug might have a similar effect in the brain, lowering chloride levels and making nerve cells responsive to GABA activation.
Staley and colleagues in Colorado conducted a trial in baby rats and found that bumetanide indeed inhibited seizure activity, while phenobarbital, as in humans, worked poorly.
