Butylated hydroxytoluene was tested for carcinogenicity in mice and rats by oral administration in the diet.
In another study in mice using one dose level and a small number of animals, the number of mice with lung tumours was increased by feeding of butylated hydroxytoluene; this finding was not confirmed in a further study by the same investigator using a larger number of animals.
Butylated hydroxytoluene was studied in mice and rats for its ability to modify the carcinogenicity of selected chemical agents.

An undergraduate experiment for the quantitative determination of the synthetic antioxidant butylated hydroxytoluene (BHT) in chewing gum is described.
Butylated hydroxyanisole (BHA) was added as a recovery surrogate, and gas chromatography-mass spectrometry with selected-ion monitoring (SIM) was used to identify and quantify BHT.
Butylated hydroxytoluene recoveries from the chewing gum matrix were typically > 80%, and student results (42-172 mg BHT per g gum) agreed closely with BHT concentrations determined in other studies (50-200 mg BHT per g gum).

This experiment then gives us an insight on how this preservative may possibly affect the human nervous system.
The hypothesis proposed that if the concentration of the preservative butylated hydroxytoluene is high, then the fruit flies exposed to the higher concentrations of BHT will have a higher average behavioral score than the flies exposed to lower concentrations of BHT.
