 About Construction of Full-Length cDNAs
Construction of full-length cDNAs is a central focus in the post-sequence era of the various genome projects, including Arabidopsis.
However, it is generally agreed upon that a cDNA comprising the entire coding sequence of a protein should be considered worthy for full-length sequencing at high accuracy.
All three investigators have constructed numerous cDNA libraries during the last 20 years in a variety of vectors that are available to the Arabidopsis community.

 Sharma, Mol Vis 2002; 8:251-258.
Approximately one thousand of the resulting subtracted cDNA clones were partially sequenced and analyzed, and a non-redundant set of one hundred of these cDNAs were examined for tissue expression pattern using a mini-Northern blot procedure and for identity by sequence analysis.
Although these cDNAs had no homology to "known genes," some bovine cDNAs exhibited significant homology to uncharacterised human cDNAs or to human genomic sequences in the database (Table 3) suggesting that latter are the human orthologues of the bovine clones.
In conclusion, all the subtracted bovine cDNAs whose expression was detectable by Northern blotting were expressed in the RPE and/or retina, demonstrating the efficacy of the subtraction method.

 DeWitt
The specific aims of the gene expression project are to (1) elucidate the metal-specificity of putative cadmium-induced cDNAs; (2) detect and sequence putative cadmium-induced cDNAs; and (3) isolate and sequence the corresponding full-length cDNAs for possible identification.
A student working on this project isolate RNA from cells under conditions of interest, design sequence-specific primers for the putative cDNAs already in hand, and use those primers to drive RT PCR reactions on the isolated RNA to determine whether or not those sequences are expressed under the various conditions.
A student working on this project would isolate RNA, use the RNA in RT-PCR reactions using degenerate primers or as template for subtractive hybridization, and isolate, amplify and clone promising new target cDNAs for sequencing to be used for future characterization studies.
