Carpe Jugulum (or "seize the throat") is exactly what the vampires -- or, rather, vampyres: as Vlad says, it's more modern -- here intend to do, and to as many of Lancre's inhabitants as possible.
Lancre's witches to save their homeland from the vampyres, who have developed new techniques to overcome many traditional vampyric weaknesses and who have decided to take over Lancre and turn it to their purposes.
The plot and subplots in Carpe Jugulum all relate to the power of belief and how it affects the world.

As with almost any Discworld novel, Carpe Jugulum starts with a select bundle of clichés, adds seasoning, a twist of the unexpected and then the author lets it rise with just a few well-concealed ingredients to spice up the mix.
In this case, the clichés he starts with are a royal christening with suitable witchy godmothers (for this is a Lancre story, which means Granny Weatherwax and her coven are in attendance) and some quickly unwelcome guests (a family of vampires from neighbouring Uberwald, invited in as a result of King Verence's political naïveté).
As a result, Carpe Jugulum is an enormously satisfying read, clever in more ways than just being very funny (which almost goes without saying).

Until a few chapters before the end you are kept in the dark (a vampire-riddled dark, scenting of blood and things profoundly evil) as to whether the witches will come out on top.
"Carpe Jugulum", IMHO, is the first Discworld novel where the plot is to thick it makes you forget that Terry Pratchett is no more as relentlessly funny as he used to be.
Which isn't to say that "Carpe Jugulum" isn't funny, no, indeed, it's a very entertaining book that will cause the occasional slap on your knee as well as an admiring nod here and there.

I love all of the Granny Weatherwax/Nanny Ogg Discworld novels, and even though "Carpe Jugulum" tackles some unusually serious themes (its vampires are truly evil, unlike the loveable, teetotalling Otto in "The Truth"), it is still vintage Pratchett and vintage Granny.
With Carpe Jugulum, Terry Pratchett returned to the witches of Lancre series.
Carpe Jugulum is already the 23rd instalment of the immensely popular Discworld series and still Terry Pratchett seems far from loosing his magical touch.
