Process to obtain oily, highly purified phosphatidylcholines - United States Patent 4,235,793
No. 1,053,299, assigned to the assignee of the present application, discloses a process to obtain natural choline phosphoric acid diglyceride esters which are free of colamin phosphoric acid diglyceride esters, by use of column chromatography where aluminum oxide is used, among other substances, as the adsorbent.
It was unexpected, and contrary to the teaching of the present state of art, that an oily alcoholic phosphatide solution would be suitable without any pre-treatment for a chromatographic separation on aluminum oxide, and where the high oil-content of the solution does not interfere in any manner with the adsorption of the acid phosphatides.
After evaporation of the solvent in vacuum under nitrogen protection, there remains 33 gram of a pliable yellowish phosphatidylcholine fraction which is free of cephalin and which can be transformed into a pourable form by the admixture of oil or partial glycerides with over 50% of unsaturated fatty acids.

The kaolin cephalin clotting time thus tests function of the intrinsic pathway, and is sensitive to isolated or combined deficiencies of factors XII, XI, X, IX, VIII, V, II, or fibrinogen.
Both the kaolin cephalin clotting time and prothrombin time are markedly prolonged, but will be corrected by the addition of normal plasma to the patient's plasma.
The development of a lupus anticoagulant is associated with prolongation of the kaolin cephalin clotting time and, in about a quarter of cases, prolongation of the prothrombin time.

This problem can be avoided, while preserving the charged polar character of the head of the molecule, if the phosphate group is esterified again with an alcohol that carries its own positive charge.
Such a molecule is ethanolamine, HO-CH which at neutral pH exists in the ionized form: HO-CH This second esterification produces phosphatidyl ethanolamine, or cephalin (shown here).
The cephalin molecule is half hydrophobic and half charged polar, yet electrostatically neutral because it carries its counter ion with it.

Using this newly developed method, he characterized the isolated "cephalin" fraction from brain tissue that after Thudichum was considered as pure phosphatidyl ethanolamine.
These research led to Folch's first publication on brain lipids in 1941 followed over a period of several yeras by a series of famous papers showing that cephalin was not a single lipid but rather a mixture of three lipids (phosphatidyl ethanolamine, serine and inositol).
After his retirement in 1977, he was Professor of Neurochemistry Emeritus and continued to be active as honorary biochemist at McLean Hospital until his death (3 October 1979).

MODIFICATIONS OF COAGULATION IN THE BURN PATIENT,
Since this protein belongs to the group of the so-called acute stage reacting proteins, its increase may be considered non-specific and as such does not indicate a hypercoagulation state, although contributing to it.
Such indirect cooperation of fibrinogen to coagulation is supported by other factors such as normal or slightly increased prothrombin activity and normal cephalin time, suggesting an activation of the coagulation chain from thromboplastic substances released from the burned surface (13, 14).
We did not observe a significant increase in FDPs, contrary to what is commonly reported in the literature (15, 16, 17), and this relates to a non-increase in plasma fibrinolytic capacity shown by our patients.
