 All About Agar
Chemically, agar is a polymer made up of subunits of the sugar galactose, and is a component of the cell walls of several species of red algae that are usually harvested in eastern Asia and California.
Although agar's chief use is as a culture medium for various microorganisms, particularly for bacteria, its other less well-known uses include serving as a thickening for soups and sauces, in jellies and ice cream, in cosmetics, for clarifying beverages, and for sizing fabrics.
Agar is a gel at room temperature, remaining firm at temperature as high as 65°C. Agar melts at approximately 85°C, a different temperature from that at which it solidifies, 32-40°C. This property is known as hysteresis.

We assessed five agar media [Sheep's blood agar (SBA), Chocolate agar (CA), Inhibitory Mold agar (IMA), Brain Heart Infusion agar (BHI) and Sabouraud-Dextrose agar (SDA)], used in conjunction with the Isolator(R) system (Wampole Laboratories, Cranbury, NJ), for the recovery of fungi from blood cultures.
Sheep's blood agar and CA were incubated for three days, primarily for the recovery of bacteria, while the standard fungal media (IMA, BHI, SDA) were incubated for 21 days.
In conclusion, chocolate agar is an excellent media for the recovery of most fungi from the Isolator(R) blood culture system; further studies investigating extended incubation of CA for the recovery of H. capsulatum should be performed.
