Class I molecules are encoded by the BCA region while class II molecules are encoded by the D region.
Class I molecules are composed of two polypeptide chains; one encoded by the BCA region and another (ß2-microglobulin) that is encoded elsewhere.
Class II molecules are composed of two polypeptide chains, both encoded by the D region.

 immunology - MHC complex
Class I MHCs are made up of one large polypeptide chain, the alpha chain which is coded for by MHC class I genes and about 45kilodaltons in weight.
MHC class II MHC class I molecules are found on almost all cells and alert the Tc cells to any infection or damage in body tissues.
Class I MHC is found on all cells and presents antigens on these cells so Tc cells must be able to recognize MHC class I to ensure that Tc cells can remove infected cells whatever or wherever they are in the body.

Major histocompatibility complex - Wikipedia, the free encyclopedia
The best-known genes in the MHC region are the subset that encodes cell-surface antigen-presenting proteins.
Loading of class II molecules must still occur inside the cell; extracellular proteins are endocytosed, digested in lysosomes, and bound by the class II MHC molecule prior to the molecule's migration to the plasma membrane.
Because class II MHC is loaded with extracellular proteins, it is mainly concerned with presentation of extracellular pathogens (for example, bacteria that might be infecting a wound or the blood).

Antigen Processing and Presentation   (Site not responding. Last check: 2007-11-06)
MHC class II molecules present fragments derived from extracellular (exogenous) proteins that are located in an intracellular compartment.
Class I MHC is a heterodimer of membrane-bound α chain and non-covalently associated β2-microglobulin.
Peptides which bind class II are from 9 to at least 30 amino acids long and are not anchored by their amino and carboxyl termini (See Figure 3).

MHC's central position in the formation of an immune response is derived from its role in presenting Ag to T-cells.
Class I and Class II molecules have similar binding sites so it is expected that the peptides they bind will also be similar in structure.

mekentosj.com | Science - MHC Class II antigen presentation
The MHC class II pathway processes material taken up by the cell from its environment into small peptides (also called antigens), which are subsequently loaded on the MHC class II molecules from which this pathway derives its name.
The MHC class I pathway is functional in all nucleated cells, and continuously samples the intracellular protein content for signs of potential danger coming from within, like viral infections or tumorigenic signals.
In contrast, the MHC class II pathway samples material that is taken up by the cell from the outside world, looking for pathogens like bacteria and parasites that live extracellularly.

Enterotoxin B Superantigen Interation with MHC Class II
The T-cell receptor binding sites on the superantigen molecules in the complex are also positioned near but not within the MHC peptide binding pockets.
These data help explain how superantigen bypasses the normal routes for specific recognition of antigen by MHC class II molecules and T cell receptors resulting in antigen-nonspecific activation of the cells expressing these receptors.
The consequence is an unusually vigorous polyclonal activation of T cells with an exaggerated immune-like response that can be life-threatening in the worst-case scenarios.
