Fourth, we have employed cell culture techniques with the A6 cell line and primary cultures of rabbit cortical collecting duct cells in an attempt to determine the regulatory mechanism which appears to prevent an effect of vasopressin on Na+ transport in the native rabbit cortical collecting duct.
In particular, we have found that after growth in primary culture cortical collecting duct cells that are immuno-dissected from the rabbit renal cortex are no longer refractory to the stimulatory effects of vasopressin, but exhibit a sustained increase in Na+ transport when the hormone is added.
We are presently conducting studies with isolated perfused cortical collecting ducts from Dahl salt-sensitive rats to determine whether the same or different hormonal regulatory mechanisms are present as in the salt-resistant rat, and in the normal Sprague-Dawley rats that we have been using in other studies.
