In vertebrates, DNA methylation typically occurs at CpG sites, where a cytosine is directly followed by a guanine in the DNA sequence.
CpG sites are often found at higher density near gene promoters where they are collectively referred to as CpG islands.
Two base-specific cleavage reactions (T reverse and C reverse) are performed to discover methylation sites and to determine methylation ratios within a given target region.

CpG sites are regions of the DNA where a cytosine nucleotide occurs next to a guanine nucleotide.
CpG sites thus tend to be eliminated from the genomes of eukaryotes.
Surprisingly, these CpG sites are unmethylated, and therefore any spontaneous deaminations of cytosine to uracil are recognized by the repair machinery and the CpG site is restored.

If CpG island prediction is not used for primer selection (default), PCR products must span a minimal number of CpG sites specified by user (default: 5).
Primers should contain at least one CpG site within their sequence, and the CpG site should preferably be located in the most 3’-end of their sequence to discriminate methylated DNA against unmethylated DNA.
If CpG island prediction is used for primer design and more than one island is found, any of the predicted island will be the target region for amplification.

I am aware that some people are interested in directly promoting CPG and some of them are a a little overzealous bordering on the questionable tactic side.
I only have one CPG site and I only had to make ONE mod to a lang-english.php file, and that was only because am thorough, the change was able to be made in the interface itself, but I knew I'd rest better if I made the change in teh lang file.
I'm not saying CPG is perfect, but my limited experience with it has proved itself as being a more capable and fitting alternative.
