Cyclooxygenase - Wikipedia, the free encyclopedia
Cyclooxygenase (COX) is an enzyme (EC 1.14.99.1) that is responsible for formation of important biological mediators called prostanoids (including prostaglandins, prostacyclin and thromboxane).
Pharmacological inhibition of COX can provide relief from the symptoms of inflammation and pain; this is the method of action of well-known drugs such as aspirin and ibuprofen.

AKS Use Case - Digging out specific COX inhibitors   (Site not responding. Last check: 2007-10-21)
Cyclooxygenases are a family of well-known enzymes that regulates the conversion of arachidonic acid to prostaglandin.
Although both cyclooxygenases share a common biological function, we can see that they have different cooccurrences with diseases and chemicals probably due to their diferent roles in the organism.
Thanks to AKS we were able to see relevant documents regarding cyclooxygenases, find the different association to diseases and drugs of the two main isozymes, establish a time-line research and look for the main authors with whom we might want to collaborate in our possible future research on cyclooxygenases.

Both forms of PGHS are heme proteins composed of two 70,000 Dalton subunits, and possess both cyclooxygenase and peroxidase activity.
Inhibition of the cyclooxygenase activity of PGHS is responsible for the anti-inflammatory activity of aspirin, indomethacin, ibuprofen, piroxicam, and other non-steroidal anti-inflammatory drugs (NSAIDs).
The committed step in the biosynthesis of the prostaglandins, thromboxanes, and prostacyclin is carried out by the membrane-bound enzyme prostaglandin G/H synthase (PGHS), which exhibits both cyclooxygenase and peroxidase activities.

The second form of cyclooxygenase, cyclooxygenase-2, is rapidly and readily inducible by a number of agents including mitogens, endotoxin, hormones, cytokines and growth factors.
As prostaglandins have physiological and pathological roles, we have concluded that the constitutive enzyme, cyclooxygenase-1, is responsible, in large part, for endogenous basal release of prostaglandins and hence is important in their physiological functions such as the maintenance of gastrointestinal integrity and renal blood flow.
For the treatment of any of these cyclooxygenase mediated diseases compounds of Formula I may be administered orally, topically, parenterally, by inhalation spray or rectally in dosage unit formulations containing conventional non-toxic pharmaceutically acceptable carriers, adjuvants and vehicles.

Unfortunately, in spite of positive experimental and clinical data, recent findings suggest that it may be unwise to separate so sharply the functions of the prostaglandins produced by the two cyclooxygenases (Table 1).
It is therefore conceivable that dual inhibition of both cyclooxygenase and 5-lipoxygenase should counteract inflammatory vascular changes as well as leukocyte induced damage, resulting in a more profound antiinflammatory action than the mere blockade of cyclooxygenase.
