Drug and Alcohol Progam   (Site not responding. Last check: 2007-10-28)
In 1995, a Coast Guard Drug and Alcohol Program Inspector (DAPI) position was established in each of the nine Coast Guard Districts in an effort to increase compliance among the marine industry in respect to the chemical testing regulations.
These DAPI positions were established in the geographical center of the applicable vessel distribution in the District.
Although it is unrealistic to expect a DAPI to visit each and every marine employer within his or her district, they are expected to respond to any questions or problems you may have regarding your chemical testing program.

DAPI - Wikipedia, the free encyclopedia
DAPI or 4',6-diamidino-2-phenylindole is a fluorescent stain that binds strongly to DNA.
DAPI's blue emission is convenient for microscopists who wish to use multiple fluorescent stains in a single sample.
Because DAPI readily enters live cells and binds tightly to DNA, it is toxic and mutagenic.

FISH guide - washes, antifade, DAPI
The solution should be stored in small aliquots (for 20-30 slides) at -20° C or -70° C. The vials should be kept as little as possible at room temperature (enough to get the required amount for the slides) and should be stored back in the freezer.
Although both methods work, the quality of the banding pattern (DAPI) and the overall quality of the staining (contrast, brightness) is higher when the counterstain is added separately from the antifade.
As general guidelines, DAPI and propidium iodide staining solutions are used at a concentration of 200 ng/ml DAPI or PI in 2xSSC or PBS.

DAPI staining   (Site not responding. Last check: 2007-10-28)
4'-6-Diamidino-2-phenylindole (DAPI) is known to form fluorescent complexes with natural double-stranded DNA, showing a fluorescence specificity for AT, AU and IC clusters.
Because of this property DAPI is a useful tool in various cytochemical investigations.
When DAPI binds to DNA, its fluorescence is strongly enhanced, what has been interpreted in terms of a highly energetic and intercalative type of interaction, but there is also evidence that DAPI binds to the minor groove, stabilized by hydrogen bonds beween DAPI and acceptor groups of AT, AU and IC base pairs.
