DNA is an extremely favorable construction medium: The sticky-ended association of DNA molecules occurs with high specificity, and it results in the formation of B-DNA, whose structure is well known.
The molecule is constructed from three segments, a duplex consisting of strands L1 and L2, a duplex consisting of strands R1 and R2, and the tetramobile junction, consisting of strands JT and JB.

In that article, Dr. Axel described a process called "co\_ transformation", where a mammalian cell is transformed with the gene of interest (introduced into the cell on one DNA construct), and, at the same tim e, is transformed with a second gene known as a "marker gene" (introduced on a second DNA construct).
The claims also claimed co\_ transformation using a single DNA construct on which the marker gene and the gene of interest were linked prior to i ncorporation into the cell.
To distinguish Axel, the applicants specifically argued that their invention involved linking of the beta interferon gene and the marker gene on a single DNA construct and that the Axel publi c ation described using two unlinked constructs, one carrying the interferon gene and one carrying the marker gene.

Transgenic mice are constructed by injecting cloned DNA into the pronucleus of fertilized mouse eggs; those eggs that survive the injection are implanted in foster females to develop to term.
In particular, the purity of the DNA construct to be injected is very important to avoid toxic effects to the embryos, and the DNA must be at the proper concentration for injection.
The DNA construct must be purified away from vector sequences.
