Dantrolene - Wikipedia, the free encyclopedia
Dantrolene sodium is a muscle relaxant that is currently the only specific and effective treatment for malignant hyperthermia.
Dantrolene can also be tried to treat malignant neuroleptic syndrome, because some patients have reacted favourably.
Dantrolene depresses excitation-contraction coupling in skeletal muscle by binding to the ryanodine receptor, and decreasing intracellular calcium concentration.

Dantrolene (additional info and facts about Dantrolene) is a muscle relaxant that works directly on the ryanodine receptor to prevent the release of calcium.
Pretreatment with dantrolene has been advocated in the past to prevent MH, but this is probably unreliable, and the long half-life of the drug may leave patients weak for extended periods.
After the widespread introduction of treatment with dantrolene (additional info and facts about dantrolene) the mortality of malignant hyperthermia fell from 80% in the 1960s to less than 10%.

REASONS FOR PERFORMING STUDY: Dantrolene sodium (Dantrium) has been used extensively for the treatment of myopathies in man and anecdotal evidence suggests it is of clinical benefit in the control of exercise-induced rhabdomyolysis (ER) in racehorses, although data to support this are currently lacking.
For each horse, the difference in change between pre- and post exercise CK values between placebo and dantrolene treatments was calculated, with positive values indicating a greater rise with placebo than with dantrolene sodium treatment.
CONCLUSIONS: The results confirmed that oral administration of dantrolene sodium, 1 h before exercise, had a statistically significant effect on reducing the difference between pre- and post exercise plasma CK levels compared with a placebo in the same animals, and preventing clinical ER in susceptible individuals.
