J&J's dapoxetine effective for premature ejaculation
The data on dapoxetine, first reported in May 2005, show that the drug was able to triple the duration of intercourse in men who had been with the same partner for at least six months and persistently suffered from premature ejaculation.
Dapoxetine is a selective serotonin reuptake inhibitor, in the same class as many of the antidepressants used to treat premature ejaculation at the moment, but is the only one to have been studied this extensively in clinical trials.
Dapoxetine is the first on-demand agent for premature ejaculation, Jon Pryor, M.D., of the University of Minnesota here, and colleagues, reported in the Sept. 9 issue of The Lancet.

The percentage of men who rated their sexual satisfaction as "good to very good" nearly doubled after treatment with both doses of dapoxetine (20.2% to 38.7% for the 30 mg dose and 22.3% to 46.5% for the 60 mg dose) compared with placebo treatment (21.6% to 24.6%).

Dapoxetine, developed by Johnson & Johnson Pharmaceutical Services, acts in much the same way as those antidepressants that increase the availability of the chemical serotonin in the brain.
Pryor called dapoxetine a "very safe" drug, which comes with a very low risk of side effects such as headache and nausea and does not appear to suppress sexual desire or carry risks such as suicidal ideation -- concerns that in recent months have prompted the FDA to issue warnings associated with some antidepressants.

Dapoxetine is a selective serotonin reuptake inhibitor (SSRI) but is slightly different from the SSRIs (such as Zoloft, Paxil, and Prozac) widely prescribed for depression and other psychiatric disorders.
Hopes that Dapoxetine would become the first drug approved for premature ejaculation dimmed last October, when the FDA sent a "not approvable" letter to the manufacturer.
In the manufacturer-funded study, Pryor and colleagues compared 30-milligram and 60-milligram doses of Dapoxetine to a placebo in roughly 2,600 men with moderate to severe premature ejaculation.

New drug for the treatment of premature ejaculation has great results
Premature ejaculation may be the most common male sexual disorder according to the American Urological Association, they estimate that PE may affect 27% to 34% of men across all age ranges, in contrast to erectile dysfunction, which is estimated to affect 10% to 12% of all men, who are usually older in age.
Jon L. Pryor, M.D., chairman and program director of the Department of Urologic Surgery at the University of Minnesota and lead investigator of the dapoxetine phase III clinical trials, says PE can be a lifelong condition experienced from the beginning of sexual activity or can develop after years of satisfactory sexual activity.
The results showed that the men taking dapoxetine 30 mg and 60 mg experienced more than a three-to- four fold increase in mean IELT compared with placebo, and IELT increased significantly with the first dose of dapoxetine, and increases in IELT were maintained over the 12-week study period.
