  United States Patent Application: 0050089840
An isolated, derivatized peptide consisting of the sequence of SEQ ID NO: 3, wherein the peptide is modified by replacing glutamic acid at position 13 with a lysine and attaching a coupling group to the lysine, or derivatized by conjugating a coupling group to a lysine added at the C-terminus.
The derivatized peptide of claim 7, wherein the coupling group is selected from the group consisting of: (a) a maleimido group; (b) a succinimidyl group; (c) a hydrazine group; and (d) a carbonyl group.
The derivatized peptide of claim 13, wherein the maleimido group is 3'-maleimidopropionate connected to the epsilon amino group of lysine by [2-(2-amino)ethoxyl]ethoxy acetic acid.

Further surprising in conjunction with the derivatization of a mixture of malto-oligosaccharides is the discovery that the change in DP profile of the mixture upon oxidation, and, it is believed, other derivatization, is smaller in conjunction with reduced malto- oligosaccharides as compared with unmodified malto- oligosaccharides.
The scope of derivatization encompassed by the invention is not contemplated to be limited, and thus, for example, the hydrogenated malto- oligosaccharides may be derivatized via oxidation, esterification, etherification, or other suitable derivatization reaction.
For example, with respect to derivatization the oxidation of the malto-oligosaccharide in one or more primary alcohol positions to form carboxylic acids, a variety of oxidation reactions are known in the art and are applicable for use in conjunction with the invention.

Patent 5098977: Methods and compositions for providing articles having improved biocompatability characteristics
The aliphatic extensions employed for derivatizing the are preferably of the type having from 12 to 22 carbon atoms, such as aliphatic hydrocarbons or acyls, in that such structures are generally found to adsorb to albumin.
The degree of derivatization, e.g., the density of derivatization in terms of the ratio of derivatized to underivatized aromatic structures, will generally be determined by the length of time the reaction is allowed to proceed.
For the step of aliphatic derivatization, it is generally necessary to perform a solvent exchange to remove the polymer from its aqueous environment and replace it with polar aprotic solvent such as dry dimethylacetamide, dimethyl sulfoxide, tetrahydrofuran or dimethyl formamide.

 Analysis of diacylglycerols
Derivatized DAG are purified by TLC on silica gel with petroleum ether/diethyl ether (75/25, v/v) as solvent system.
The Rf of derivatized alkenyl-, alkylacyl- and diacyl-DAG are 0.82, 0.75 and 0.67, respectively.
The same derivatization procedure is used for the analysis of the molecular species of free diradylglycerols or of phospholipids after their hydrolysis with phospholipase C. Each sub-class, purified by TLC, is analyzed by HPLC.

A high-performance liquid chromatographic assay for the determination of desbutylhalofantrine enantiomers in rat plasma
For the nonstereospecific assay of DHF used in the determination of recovery and derivatization yield, the mobile phase consisted of a 25:75 mixture of [25 mM potassium phosphate:3 mM sulfuric acid:3.6 mM triethylamine]:acetonitrile containing sodium dodecyl sulfate (1.5 g/L).
The derivatization yield of DHF was estimated by calculating the difference in the mean underivatized DHT:HF peak height ratios between the derivatized and underivatized samples.
We chose to use the latter conditions for derivatization, since a.) the reaction was faster, b.) the peaks at 4 and 6 min eluted at a time where potentially interfering peaks from endogenous were co-eluting, and c.) heat was not required which could potentially lead to inadvertent racemization of the DHF enantiomers.

When the derivatized protein is precipitated or crystallized, the rate at which the insulin derivative dissolves from the solid form is greatly reduced compared with the rate at which similar solid forms comprised of un-derivatized protein dissolve.
Accordingly, in its broadest aspect, the present invention provides insoluble compositions comprising a derivatized protein selected from the group consisting of insulin derivatives, insulin analog derivatives, and proinsulin derivatives, wherein the derivatives are less soluble than the underivatized insulin, insulin analog, or proinsulin.
Also part of the present invention are amorphous precipitates, comprising, in their broadest aspect, a derivatized protein selected from the group consisting of derivatized insulin, derivatized insulin analog, and derivatized proinsulin, protamine, a phenolic preservative, and zinc, wherein the derivatized protein is less soluble than the underivatized protein.

Accordingly, in its broadest aspect, the present invention provides insoluble compositions comprising a derivatized protein selected from the group consisting of insulin derivatives, insulin analog derivatives, and proinsulin derivatives, a protein selected from the group consisting of insulin, insulin analogs, and proinsulins, a complexing compound, a hexamer-stabilizing compound, and a divalent metal cation.
The derivatized protein is either less soluble in an aqueous solvent than is the un-derivatized protein, is more lipophilic than un-derivatized insulin, or produces a complex with zinc and protamine that is less soluble than the corresponding complex with the un-derivatized protein.
Hybrid hexamers are believed to be formed when a protein and a derivatized protein are first mixed together under conditions that strongly favor dissolution into lower states of aggregation than the hexameric state, and second, the conditions are changed to strongly favor the hexameric aggregation state.

 Amino Acid Analysis
Derivatization is performed automatically on the amino acid analyzer by reacting the free amino acids, under basic conditions, with phenylisothiocyanate (PITC) to produce phenylthiocarbamyl (PTC) amino acid derivatives (see Figure 1 and Figure 2).
Following derivatization, a methanol solution containing the PTC-amino acids is transferred to a narrow bore HPLC system for separation.
The PTC-amino acids are separated on a reverse phase C18 silica column and the PTC chromophore is detected at 254 nm.

Distinguishing amphetamine, methamphetamine and 3,4-methylenedioxymethamphetamine from other sympathomimetic amines ...
Misidentification of ephedrine and pseudoephedrine as methamphetamine has been reported because of similar retention times of their derivatives in gas chromatography as well as very similar mass spectral fragmentation patterns in the conventional electron impact mode of analysis.
The derivatization is easily accomplished at room temperature by adding the derivatizing reagent in the extraction solvent because the reagent is stable in the presence of water.
The electron impact mass spectrum of derivatized methamphetamine (base peak, m/z 144, other peaks at m/z 102, 58) is similar to the electron impact mass spectrum of both derivatized pseudoephedrine (base peak, m/z 144, other peaks, m/z 102, 58), and ephedrine (base peak, m/z 144, other peaks, m/z 102, 58).

We review the results of spectral studies of the ultrafast excited-state absorption in fullerenes and derivatized fullerenes, which allow determination of both the spectral response of reverse saturable absorption (RSA) nonlinearities in these compounds, as well as the dynamical response for different morphologies.
We find that by appropriate pretreatment, the optical limiting response of derivatized fullerene sol-gels can be enhanced to approach that of the same molecules in solution while significantly enhancing the damage threshold.
The ultrafast absorption and optical limiting of these derivatived fullerenes will be compared with that of various porphyrin and phthalocyanine molecules as well as new broadband limiting materials which consist of phthalocyanines derivatized directly onto the fullerene molecule.

Examination of the amino acid derivatized diacetylene lipids showed a wide variety of surface characteristics with many novel properties.
Decrease of pH results in attenuation of LE–LC phase transition in Gln-PDA monolayers due to charge screening, while temperature affects the monolayers through regular thermal agitation, and the effect is opposite to the thermal anomaly observed for the non-derivatized diacetylene lipids.
EFM studies reveal that only the lipids derivatized with hydrophilic headgroups exhibit phase transition and domain formation, while those with hydrophobic headgroups do not easily form polymerizable matrices.
