		The Dicer enzyme is able to snip a double-stranded form of RNA into segments that can attach themselves to genes and block their activity.
		With this crystal structure, the researchers learned that Dicer serves as a molecular ruler, with a clamp at one end and a cleaver at the other end a set distance away, that produces RNA fragments of an ideal size for gene-silencing.
		The Dicer enzyme is known to play a key role in RNA interference, but the details of how it functions have remained unclear.

		Doudna and her colleagues solved the structure of a Dicer enzyme obtained from the parasite Giardia intestinalis using x-ray crystallography, a technique that enables researchers to construct pictures of biological molecules in superb three-dimensional detail.
		In the lab, Dicer is used in mice to switch off any combination of genes -- either in targeted or in random fashion -- to infer a gene's function, a capability that may be especially useful in understanding cancer and developing improved cancer therapies.
		Dicer enzymes are found in all cells of higher animals, suggesting that it has an ancient evolutionary heritage.

		The structure of Dicer is an elongated molecule with a PAZ domain on one end and dual RNase III domains on the opposing end.
		Dicer works by recognizing the ends of dsRNA with its PAZ domain and then cutting the dsRNA with its RNase III domains.
		The distance between the PAZ and RNase III domains determines the length of the siRNA produced by Dicer, which varies by species but is typically in the range of 22 to 30 nucleotides.
