Sweet Clover Poisoning: Introduction - The Merck Veterinary Manual
When toxic hay or silage is consumed, hypoprothrombinemia results, presumably because dicumarol combines with the proenzyme required for synthesis of prothrombin (by preventing formation of the active enzyme).
The time between consumption of toxic sweet clover and appearance of clinical disease varies greatly and depends on the dicumarol content of the particular sweet clover variety being fed, age of the animals, and the amount of feed consumed.
If the dicumarol content of the ration is low or variable, animals may consume it for months before signs of disease appear.

DICUMAROL is a blood thinner used to keep blood flowing smoothly and prevent the formation of blood clots.
Before taking DICUMAROL, tell your doctor your medical history, especially of: bleeding problems (ulcers or lengthy or heavy menstrual periods), diabetes, liver or kidney disease, high blood pressure, arthritis, thyroid problems, any infection, recent surgery, any allergies.
It is important that all doctors and dentists treating you know that you take DICUMAROL so that they can avoid prescribing other drugs that may interfere with its effect.

Pb Side Effects, and Drug Interactions - Phenobarbital - RxList Monographs
Anticoagulants: Phenobarbital lowers the plasma levels of dicumarol (name previously used: bishydorxycoumarin) and causes a decrease in anticoagulant activity as measured by the prothrombin time.
Phenobarbital can induce hepatic microsomal enzymes resulting in increased metabolism and decreased anticoagulant response of oral anticoagulants (e.g., warfarin, acenocournarol, dicumarol, and phenprocoumon).
Patients stabilized on anticoagulant therapy may require dosage adjustments if phenobarbital is added to or withdrawn from their dosage regimen.

 Blood Relations
By disrupting the balance of clotting factors in the bloodstream, large quantities of dicumarol were deemed the culprit in the cattle hemorrhaging.
This discovery paved the way for the creation of warfarin—brand name Coumadin—a derivative of dicumarol and currently one of the most widely prescribed drugs in the United States.
For the last 40 years, warfarin has been used safely and effectively to prevent stroke and treat deep vein thrombosis (clots in the legs), pulmonary embolism (clots in the lungs) and other disorders related to abnormal clot formation.

April 2003 Medical Update Mayo Clinic in Jacksonville, Fla.   (Site not responding. Last check: 2007-10-09)
Warfarin and dicumarol are vitamin K antagonists that interfere with the gamma-carboxylation of vitamin K-dependent clotting proteins.
They inhibit synthesis of factors II, VII, IX and X as well as proteins C and S. Dicumarol was introduced in 1942; warfarin in 1953.
The disadvantages of warfarin are that it requires initial overlap with heparin, has a narrow therapeutic index and requires frequent monitoring for food/drug interactions, the impact of concomitant illnesses such as liver disease and serendipitous changes in the International Normalized Ratio (INR).

Novel Plastic Beads Enhance Drug Delivery
"The exciting thing is that with a similar delivery system, we were successful in delivering a small molecule such as dicumarol (an anticoagulant); proteins, such as insulin; and also huge molecules such as DNA," said Dr. Mathiowitz, lead author on the paper.
In their current study, insulin delivered in microspheres lowered blood sugar levels within two hours, dicumarol remained for in the bloodstream up to three days, and DNA was incorporated into cells and produced a new protein within five days.
The researchers believe that the success of their work is partially due to the small size of their beads, which ranged from 0.1 to 5 microns in diameter.
