  dnaC -
Replication of bacteriophage phiK duplex replicative-form DNA in dnaB and dnaC mutants of Escherichia coli.
The ratio of the dnaC and dnaB polypeptides in the complex is approximately 1, as determined on a denaturing gel.
Kinetic analysis of the ATPase activity of dnaB X dnaC complex indicates that the decrease in the ATPase activity on complex formation is due to a reduction of the maximal velocity (Vmax).

DnaB is involved in both the initiation and the elongation stages of DNA replication, and it plays a key role in the replication of bacterial chromosomal, phage, and plasmid DNA (Kornberg and Baker, 1991).
DnaB protein was separated from DnaC and the DnaB.DnaC complex by chromatography on a column of DEAE-cellulose (Kobori and Kornberg, 1982), and further purified by gel filtration through Sephacryl S-400 (Pharmacia) in Buffer A (50 mM Tris.HCl, pH 7.6, 2 mM dithiothreitol, 5 mM MgCl2, 0.1 mM ATP, 200 mM NaCl, 20% (v/v) glycerol).
The planes corresponding to the reconstructed DnaB oligomer are presented in Figure 5, and Figure 6a and Figure 6b show two orientations of a surface rendering of the volume.

The DnaC helicase loader is a dual ATP/ADP switch protein
The DnaB mutant is inactive as a helicase, and thus the observed bubble expansion is not due to helicase action.
Normally, DnaC is not stuck tightly to DnaB loaded onto DNA and readily dissociates during gel filtration, freeing DnaB for helicase action (Kobori and Kornberg, 1982; Funnell et al., 1987; Fang et al., 1999).

dnaB protein of Escherichia coli is an essential replication protein.
By contrast, it was not inhibitory to dnaB protein in priming of single-stranded DNA by primase in the absence of single-stranded DNA binding protein.
Biochemical analysis of the altered form of dnaB protein revealed that it was inactive in replication in several purified enzyme systems which involve specific and nonspecific primer formation on single-stranded DNAs, and in replication of plasmids containing the E. coli chromosomal origin.

New intein in Cyanobacteria   (Site not responding. Last check: 2007-10-20)
The two DnaB inteins could be the result of a single integration event into a common ancestor of the cyanobacterial and chloroplast dnaB genes.
Selection on the DnaB helicases is probably linked to changes in the proteins with which they interact and to the evolution of the whole DNA replication process.
The two DnaB proteins can be aligned end to end (415 amino acids) with only 3 gaps, 37% amino acid identities and 79% amino acid identities and conserved substitutions.

 OhioLINK ETD: Shrimakar, Paresh
The dnaB gene of E.coli encodes an essential DNA replication enzyme.
In addition, general priming reactions with wild-type DnaB or RC231 in combination with ATP or ATPγS exhibited striking differences in both the efficiency of priming and the length distribution of pr imers.
Analysis of these effects suggests that primase interacts directly with DnaB throughout primer synthesis until ATP hydrolysis by DnaB disrupts the interaction and leads to primer termination.

Pfam 18.0 : DnaB   (Site not responding. Last check: 2007-10-20)
The hexameric helicase DnaB unwinds the DNA duplex at the Escherichia coli chromosome replication fork.
Although the mechanism by which DnaB both couples ATP hydrolysis to translocation along DNA and denatures the duplex is unknown, a change in the quaternary structure of the protein involving dimerisation of the N-terminal domain has been observed and may occur during the enzymatic cycle.
This N-terminal domain is required both for interaction with other proteins in the primosome and for DnaB helicase activity [1].
