We were able to reconstitute the replication with purified proteins including DnaA, DnaB, DnaC, primase, SSB and DNA polymerase III holoenzyme (10).
The hydrolysis of the nucleotide is required also to facilitate the release of DnaC protein from DnaB-DnaC complex (21).
Helicase activity of the ABC preprimosome is maximally activated by 100 to 200 µM ATP, whereas more than 1 mM of ATP is required for maximum activation of the helicase activity by DnaB protein alone (23).

DNAC is a new network access control technology designed to fill the void that current NAC solutions have yet to solve with an entirely new model that is secure, rapidly deployed, and highly cost-effective.
DNAC enforcers are unique because they quarantine noncompliant endpoints from the rest of the network, even from those PCs and servers not running the DNAC software.
DNAC's revolutionary technology ensures that rogue endpoints and guests are managed without administrator action and includes detailed reporting functions that help to assist companies in passing compliance audits and regulations.

InfoExpress - InfoExpress to Demo NAC Integration
DNAC is an innovative new technology that fills the gap between infrastructure NAC solutions which require significant network changes and updates, and other NAC solutions which are easily bypassed.
The dynamic groups of enforcers make DNAC easy to deploy, scale from small offices to large enterprise networks, and support managed and unmanaged switches and wireless access points.
DNAC does not require infrastructure upgrades, network configuration changes, or adding more network infrastructure.

DNA computing (DNAC) refers to the use of biological molecules, primarily deoxyribonucleic acid (DNA) and ribonucleic acid (RNA), for computational purposes.
A computation with biological molecules is carried out by using enzymes and techniques from molecular biology to apply a sequence of modifications to molecules in a test tube.
DNAC devices don’t appear to be suitable for applications involving simple problems or requiring quick responses, but they do hold promise for tackling large problems that are well beyond the reach of existing supercomputers.

With DNAC, the idea is it dynamically creates a pack of endpoint enforcers on the network that look for unauthorized endpoints and, when they spot them, quarantine them.
According to Lum, DNAC provides more scalability than infrastructure-based NAC because the more endpoints there are on the network, then the more enforcers that could be on the network itself.
InfoExpress' DNAC product organizes endpoints into communities of enforcers, reserves that can become enforcers, guests that don't act as enforcers and unhealthy endpoints that need to be quarantined by enforcers until they are healthy (and once they are healthy, they could become enforcers).

 Neighborhood Watch for Network Security?
The privately held InfoExpress is showing off its DNAC approach this week at the Interop networking conference in Las Vegas this week.
Instead of a hardware appliance to control NAC functions and enforcement, as is the case with vendor solutions from Cisco, Juniper and others, DNAC utilizes the collective power of a network of "enforcer" PCs to provide NAC policy enforcement.
DNAC is not without its own barriers to adoption though.

The DnaC helicase loader is a dual ATP/ADP switch protein
One might expect that ATP is required for helicase loading; however, this study on DnaC illustrates that ATP is not actually needed for DnaC to load helicase onto single-strand DNA (ssDNA).
In addition, ATP is required for DnaC function at an early step in oriC replication in which ATP stimulates ssDNA binding by DnaC, leading to expansion of the ssDNA bubble at the origin.
These observations have led to the idea that DnaC is a 'dual' switch protein, where both the ATP and the ADP forms are sequentially required for replication.

The structure of the nucleotide-binding site of the Escherichia coli replication factor DnaC protein and the effect of the nucleotide cofactor on the protein structure have been examined using ultraviolet, steady-state, and time-dependent fluorescence spectroscopy.
However, in spite of a large spatial separation, binding of both cofactors induces significant and different changes in the structure of the environment of tryptophans, indicating long-range structural effects through the DnaC molecule.
Moreover, only ATP induces changes in the distribution of the tyrosine residues on the surface of the protein.
