In the rat, the mediodorsal nucleus is thought to be defined by its connections with the prefrontal region, although the lateral part of this nucleus projects to the anterior cingulate and secondary motor areas [95][144].
It is bordered dorsally by the paratenial nucleus, ventrally by the anterior half of the hypothalamic paraventricular nucleus, and rostrally and laterally by the fornix and bed nucleus of the stria medullaris [35][225].
The endbrain projections of the intermediodorsal nucleus reportedly [23][97] are similar to those of the paraventricular nucleus because this nucleus projects to the infralimbic and agranular insular areas, although its projections are denser in the agranular insular area than those from the paraventricular nucleus, and lighter in the infralimbic area.

The nuclei investigated were the hypoglossal nucleus, dorsomedial nucleus of the intercollicular midbrain, central nucleus of the archistriatum, central nucleus of the lateral neostriatum, oval nucleus of the hyperstriatum ventrale, medial division of the oval nucleus of the anterior neostriatum, and magnocellular nucleus of the lobus parolfactorius.
At hatching the hypoglossal nucleus exhibits adult-like cytoarchitecture, and the central nucleus of the archistriatum and the central nucleus of the lateral neostriatum are distinguishable from surrounding fields.

An area called the PVH (paraventricular hypothalamic nucleus) probably uses its connections to the pituitary gland and the autonomic nervous system to execute the brain's responses to leptin.
The researchers suggest that the dorsomedial hypothalamic nucleus serves as a gatekeeper controlling the flow of information to the PVH, like an assistant outside the chief executive's office.
It then sends its output--possibly orders to change certain hormone levels--to the dorsomedial hypothalamic nucleus gatekeeper, which forwards the message to the PVH for the order to be executed through contacts at the pituitary gland.

When GLP-1 is given centrally to naïve rats it results in marked induction of c-Fos protein in the supraoptic, paraventricular nucleus and amygdala but only a moderate increase in the arcuate nucleus.
Experiments using arcuate nucleus ablated rats (MSG-rats) have shown that the GLP-1 feeding effect is likely mediated via interaction with neurons in the arcuate nucleus.

Within the TH-ir neurones, PRL induced a significant increase in c-Fos in the dorsomedial portion of the mid-arcuate nucleus (p < 0.05).
PRL also induced c-Fos expression in the supraoptic nucleus (SON) (11.7 ± 3.2 vs. 3.0 ± 1.4 cells/section for PRL and control rats, respectively; p < 0.05), but not in the medial preoptic nucleus, ventromedial nucleus or the dorsomedial nucleus, areas reported to either contain gonadotropin-releasing hormone neurones or express PRL receptors.
While the role of PRL-responsive neurones in the SON remains to be elucidated, these findings support the notion that the central actions of PRL could be mediated via the TH neurones of the dorsomedial arcuate nucleus and/or by a population of neurones in the rostral arcuate nucleus that contain neither TH nor POMC.
