Electrospray ionization - Wikipedia, the free encyclopedia
The invention of electrospray ionization was rewarded with the attribution of the Nobel Prize in Chemistry to John Fenn in 2002.
Electrospray ionization is the primary ion source used in liquid chromatography-mass spectrometry.
The use of the word "ionization" in "electrospray ionization" is criticized by some due to that many of the ions observed are thought to be preformed in solution before the electrospray process or created by simple changes in concentrations as the aerosolized droplets shrink.

 Introduction to Mass Spectrometry
Electrospray ionization generates ions directly from solution (usually an aqueous or aqueous/organic solvent system) by creating a fine spray of highly charged droplets in the presence of a strong electric field (typically 3.5 kV).
The FAB ionization technique is a soft ionization method that typically requires the use of a direct insertion probe for sample introduction and a high energy beam of xenon atoms or cesium ions to sputter the sample and matrix from the probe's surface.
While electron ionization is one of the most widely used methods of ionization in mass spectrometry, the principal problems associated with it include excessive fragmentation in the ionization source, the involatility of large molecules and thermal decomposition during vaporization.

Electrospray ionization mass spectrometry (ESI-MS) is particularly useful for very high molecular weight biopolymers such as proteins and nucleic acids greater than 10 kDa in mass, for it affords a distribution of multiply-charged molecules of the sample biopolymer without causing any significant amount of fragmentation.
Electrospray ionization produces several multiply charged ions of the parent nucleic acid, without any significant fragmentation of the nucleic acid.

Chemical Principles of Electrospray Ionization Mass Spectroscopy
Electrospray Ionization uses an electric field to produce a spray of fine droplets, which is used to create the spectrum.
Ions, residual droplets, and vapor formed by electrospray are extracted through a small hole into two more evaporation chambers (evacuated) via a “nozzle” and a “skimmer,” which then passes into the analyzer of the mass spectrometer where a mass spectrum of the original sample is obtained.
Even more attractive is the tendency for electrospray to produce multi-charged ions, a benefit that makes accurate measurement of large relative molecular masses much easier.

While electrospray ionization is a popular technique for the analysis of a wide range of compound classes, less polar and neutral compounds may require an alternative approach, such as APCI or APPI.
Using electrospray, a strong electric field is applied to the liquid sample stream, which is then nebulized and desolvated with the assistance of high temperature gas flows to produce gas phase ions.
Many laboratories, particularly in drug discovery, are tasked with analyzing compounds with a wide range of chemical structures and properties.

 Electrospray
Electrospray ionization (ESI) is an ionization technique for small amounts of large and/or labile molecules such as peptide's, proteins, organometallics, and polymers.
ESI is a soft ionization technique and is particularly useful for obtaining the molecular weight of analytes that would normally exceed the normal mass range of sector and quadrupole instruments.
The most obvious feature of an ESI spectrum is that the ions carry multiple charges, which reduces their mass-to-charge ratio compared to a singly charged species.

Soon after the introduction of TSP, the electrospray ionization (ESI) source was presented by Yamashita and Fenn
Positive or negative ions can be produced, but a stable negative ion current is more difficult to maintain due to an increased propensity toward electric discharge at the electrospray needle tip.
ESI differs from TSP in the fact that it uses a high potential to impose a charge in place of the buffers used in TSP.
