Enzyme-linked immunosorbent assay (ELISA) for determining anti-bodies against herpes simplex virus (HSV) types 1 and 2 ...
A preferred embodiment of the method employs a variation of the enzyme-linked immunosorbent assay (ELISA) whereby monoclonal antibodies are used to purify target antigens, and test sera are absorbed with virus-infected cell extracts to remove intertypic cross-reacting antibodies.
For assaying antibodies to HSV-2, one serum sample is mixed with an equal voume (100 ul) of HSV-1 infected cell extract diluted 1:10 with BSA-PBS, and a duplicate sample is mixed with an equal volume of uninfected Vero cell extract diluted 1:10 with BSA-PBS.

 ENZYME-LINKED IMMUNOABSORBENT ASSAY
The enzyme-linked immunoabsorbent assay is serologic test used as a general screening tool for the detection of antibodies to the HIV virus.
ELISA technology links an a measurable enzyme to either an antigen or antibody.
In this way, it can then measure the presence of an antibody, or an antigen in the bloodstream.

AEGiS-11IAC: HIV-specific IgA antibodies in vertically exposed babies.
STUDY DESIGN: A modified IgA antibody-capture enzyme -linked immunoabsorbent assay was used to detect HIV-specific IgA antibodies in serial plasma samples collected from 65 infants born to HIV-infected mothers in the Bahamas and in Toronto.
The specificity of this assay when evaluated for babies born to seronegative women is 100%.
Loss of these antibodies in 100% of uninfected babies after 8 months of age and its continued presence in infected babies makes this test useful in the diagnosis of HIV infection in areas where other serological tests are not reliable.

(2) It is still unknown whether the use of the FFN assay results in a decrease in the rate of morbidity and mortality related to preterm birth.
However, observational studies in women with symptoms suggestive of PTL report that the positive predictive value of the FFN assay for preterm birth ranged from 0.45 to 0.81, while the negative predictive value for preterm birth within 14 days of the test was 0.95 to 0.99.
Although the FFN assay is a risk predictor of preterm delivery among asymptomatic women, there is no proven effective treatment for asymptomatic women at risk of preterm birth that is known to improve maternal or fetal outcomes.

 DEFRA Science
Currently there is no internationally approved enzyme linked immunoabsorbent assay (ELISA) which can be used to test companion animal sera for the presence of rabies virus specific antibodies in response to vaccination.
the main obstacle to the development of such an assay is the production of an appropriate cost-effective antigen which does not require culture of live rabies virus.
The prototype assay will be validated against a large panel of post-vaccination sera and against the fluorescent antibody virus neutralisation (FAVN) test, the current Office International des Epizooties (OIE) gold standard.

The traditional latex agglutination assay has fallen into disuse because of its lower sensitivity compared to other methods; turbidimetric techniques and enzyme-linked immunoabsorbent assay (ELISA) techniques are the most sensitive, whereas the usefulness of the SimpliRED assay is still under debate.
A prospective study of consecutive patients attended between September 2002 and July 2003 in the Emergency Room of the Hospital de la Princesa in Madrid with clinical suspicion of pulmonary embolism was conducted.
Semiquantitative latex agglutination assays are rapid, simple, and cheaper, but sensitivity is limited (83%) and the negative predictive value for pulmonary embolism is 90 %.

The protocol requires a high concentration of protease inhibitor in the extraction buffer; the use of antiserum cross-absorbed with control tissue, an increased concentration of blocking reagents in the antiserum buffer, and performing all coating and/or binding processes at 37°C while reducing the time period for each step to 1 h.
