The present study was designed to characterise the acute effects of ethanol on the metabolism of ethylmorphine and to compare it with the effects on codeine in suspensions of freshly isolated rat hepatocytes.
Ethylmorphine, codeine and their metabolites were quantified by HPLC with UV detection.
The ratios between reaction rate constants in the ethylmorphine experiments at 100 and 0 mM ethanol were 0.65 for ethylmorphine-->norethylmorphine, 0.63 for norethylmorphine-->normorphine, 0.56 for ethylmorphine-->morphine, 0.49 for morphine-->normorphine, 0.31 for normorphine-->normorphine-3-glucuronide and 0.49 for morphine-->morphine-3-glucuronide.

Age related response of di(2-ethylhexyl)phthalate (DEHP) was studied by administering the plasticizer (2000 mg/kg, orally) to 3, 6 and 10 week old wistar rats for 1, 7 or 15 days and determining the activity of hepatic P450 monooxygenases.
Single administration of DEHP decreased the cytochrome P450 contents and activity of arylhydrocarbon hydroxylase, aniline hydroxylase and ethylmorphine N-demethylase at all the age groups while repeated exposure induced them with maximum alterations occurring in 3 week old rats.
Repeated administration for 15 days, on the other hand, caused a decrease in the cytochrome P450 content and the activity of arylhydrocarbon hydroxylase, aniline hydroxylase and ethylmorphine N-demethylase only in the 3 week old rats.

Joseph von Mering, a physician who was to become famous for his discovery of the hypnotic barbiturates, included the drug among 18 morphine derivatives that he tested for E Merck of Darmstadt.
von Mering was particularly impressed with ethylmorphine, a compound synthesized by Grimaux in 1881,which he found to be efficacious in the alleviation of coughs, even in patients with pulmonary tuberculosis in whom codeine had proven ineffective.
Ethylmorphine was the first semi-synthetic morphine derivative to be introduced into the clinic and probably influenced the subsequent decision of F Bayer and Co to introduce heroin for similar clinical purposes.

The initial temperature was 150° C for 1 min and was then increased with the rate of 15° C per min to 220° C and, finally, with the rate of 10° C/min to 320° C where it was held for 1 min.
The ions followed were m/z 357 and 284 for dihydrocodeine (RT 9.9), m/z 282 and 355 for codeine (RT 10.2), m/z 296 and 369 for ethylmorphine (RT 10.4), m/z 327 and 268 for 6-acetylmorphine (RT 11.1) as well as m/z 341 and 324 for morphine (RT 11.7).

Approved indication for dihydrocodeine is the management of moderate to moderately severe pain as well as coughing and shortness of breath.

7-Ethoxyresorufin (EROD), 7-pentoxyresorufin (PROD), aniline and ethylmorphine were used as substrates for CYP 1A, CYP 2B, CYP 2E1 and CYP 3A, respectively.
Urethane increased EROD (CYP 1A) activity by 40 % (p < 0.01), and hydroxylation of aniline (CYP 2E1) by 14 % in the early phase of anaesthesia and by 60 % (p < 0.01) in the later one.

It was found that during incubation of separate alkaloids of chelidonium (berberine, chelidonine, chelerythrine, sanquinarine and protopine) with microsomal fraction of liver rats in the presence of oxygen and NADPH2 the formation of formaldehyde was noted, which presents evidence of O- and N-demethylation of alkaloids.
A positive correlation was shown between the activity of sanquinarine-N-demethylase and the velocity of dealkylation of ethylmorphine (cytochrome P450 3A-dependent reaction), aminopyrine (cytochrome P450 2C11-dependent reaction) and total content of cytochrome P450 in liver microsomal fraction.
The administration of alkaloids to rats (for 10 days at doses of 5, 10 and 20mg/kg/day) caused activation of cytochrome P450-dependent liver microsomal monooxygenases.

The data presented here document a protective effect of dietary methyl donors on AFB1-induced hepatocarcinogenesis in mice probably acting, in part, via activation/detoxification mechanisms favoring an increased balance in detoxification of AFB1.
