Topic: Etoperidone

Patent 4132791: Use of etoperidone in parkinsonism and in other extrapyramidal syndromes characterized by tremors
The drug etoperidone is presently being used in therapy as a psychotropic agent, the main field of application being primary or secondary depressive and anxious-depressive states.
Clonidine-induced tremors are inhibited by trazodone and etoperidone.
Etoperidone was studied as follows: a double blind placebo controlled cross-over clinical trial was performed on a group of 10 patients (7 males and 3 females) suffering from Parkinsonism and ranging in age from 60 to 72 years (average age: 64).

Patent 4131675: Use of combinations of L-DOPA with trazodone and L-DOPA with etoperidone in Parkinsonism
A combination of trazodone or etoperidone with L-DOPA is used in the treatment of Parkinsonism by oral administration to a patient.
The effects of trazodone and etoperidone on the signs of adrenergic stimulation produced by L-DOPA were studied by considering piloerection and mydriasis which represent the most typical expressions of adrenergic stimulation.
Trazodone and etoperidone were administered at a fixed dose of 50 mg/kg.

Blind- ability takes on added importance, in the light of recent compelling reports which estimate that up 70% of patients affected with mild medical condi- tions respond very favorably to placebo (Goleman, 1993).
White, Kando, Park, Waternaux, and Brown (1992) retrospectively assessed the "blindability" of a clinical drug trial of etoperidone, a putative antidepressant.
An evaluator was provided with all the drug trial data minus the outcome results.

I3C's potential as a chemopreventive agent was confirmed in a study funded by the National Cancer Institute where 90 different cancer preventatives were put through six different tests of cancer prevention.
I3C was one of 8 compounds proven effective in all six assays (the other 7 were ascorbic acid, bismuththiol, esculetin, etoperidone, folic acid, hydrocortisone and tocopherol succinate).
Another potential action of I3C against chemical cancers is its potential to keep dioxin and other chlorine chemicals out of cells.

Synergistic Action of 5-HT2A Antagonists and Selective Serotonin Reuptake Inhibitors in Neuropsychiatric Disorders   (Site not responding. Last check: 2007-11-05)
In addition to the well-known 'atypical' antidepressants listed above, there are double-blind studies demonstrating antidepressant activity of other potent 5-HT antagonists as well, such as ritanserin, pipamperone, pizotifen, danitracen, and etoperidone (Matussek et al, 1976; Ansoms et al, 1977; Standal, 1977; Kern and Richter, 1985; Bersani et al, 1990).
In contrast to the preliminary evidence that blockade of 5-HT receptors may augment the therapeutic effects of SSRIs in OCD, evidence from monotherapy with potent 5-HT antagonists in this disorder is generally negative.

Thioridazine was also associated with significantly more dizziness.
No superiority for thioridazine was shown in comparisons with etoperidone, loxapine or zuclopenthixol, except to produce fewer side effects than loxapine.
Reviewers' conclusions: Very limited data are available to support the use of thioridazine in the treatment of dementia.
