FAS ligand - Wikipedia, the free encyclopedia
The FAS ligand is a type II transmembrane protein that belongs to the tumor necrosis factor (TNF) family.
FAS is a protein which may bind to the surface of a cell possibly leading to apoptosis, or cell death.
Germline mutation of Fas is asociated with Autoimmune Lymphoproliferative Syndrome (ALPS) (OMIM #601859), a childhood disorder of apoptosis.

Strong Fas expression was correlated with depth of invasion (40.3% in pT1,T2 versus 20.5% in pT3,T4; P = 0.0308), histological differentiation (45.7% in well versus 25.4% in nonwell; P = 0.0347), and lymph node metastasis (22.6% in positive versus 45.5% in negative; P = 0.0129).
Fas expression was one of the independent favorable prognosticators for patients' survival (risk ratio, 3.26; P = 0.0103) in esophageal SCC.
Fas expression was an independent prognosticator for recurrence-free survival, whereas FasL expression did not influence the survival in esophageal squamous cell carcinoma.

The status of Fas and Fas ligand expression can predict recurrence of hepatocellular carcinoma   (Site not responding. Last check: 2007-11-02)
The status of Fas and Fas ligand (Fas L) expression was investigated in this study for 103 hepatocellular carcinomas (HCC).
Fas L expression in carcinoma cells was observed exclusively in moderately or poorly differentiated cases.
These results suggest that the evaluation of Fas and Fas ligand expression potentially has a significant prognostic value for DFS of HCC patients, in addition to the clinical stage, and can be regarded as a new prognostic marker.

Activity: Reacts with Fas Ligand (FAS-L), a 40 KD type II integral membrane protein, homologous with tumor necrosis factor (TNF) and expressed on activated T and NK cells.HUman FASL is released from a 40Kd molecule as a 26KD soluble protein from transfectant cells or activated T cells by matrix metalloproteinases (MMPs).
Fas L is a tyep II transmembrane protein which binds to its counter- receptor CD95 (FAS) and is speculated to participate in regulation of a variety of immunological responses and lymphocyte development.

Functional Fas-ligand expression on T cells from HIV-1-infected patients is unrelated to CD4+ lymphopenia.
Recent studies have demonstrated that the expression of Fas by peripheral T cells from HIV-1+ patients is deregulated and increases the susceptibility of these cells to undergo apoptosis.
Since CD4+ cells of Th1 phenotype are impaired during HIV-1 infection and show high cellular expression of Fas-L, it is conceivable that excess Fas-L during the early or non-lymphopenic phase of the disease increases the extent of apoptosis in these cells by the Fas/Fas-L pathway.
The defective expression of the ligand in severely lymphopenic stages could be explained by exhaustion of this mechanism as the disease progresses.

Fas Ligand (FasL) has been shown to mediate both apoptotic and inflammatory reactions.
When FasL binds to Fas, it oligomerizes the trimeric receptor that results in the recruitment of the death-inducing signal complex.
Considerable confusion exists in the literature as to the relative importance of the membrane and soluble form of FasL on the proinflammatory effects of this molecule.

In this study, we hypothesized that soluble form of Fas (sFas) and FasL (sFasL) may also be associated with this disorder.
Molecular cloning and nucleotide sequence analysis have revealed a human Fas messenger RNA variant that encodes a soluble Fas (sFas) molecule lacking the transmembrane domain because of the deletion of an exon encoding this region.[10] sFas blocks apoptosis in cells by inhibiting the binding of FasL to Fas on the cell membrane.[10,11]
We previously demonstrated that the expression of Fas was upregulated in bronchiolar and alveolar epithelial cells and that FasL protein was upregulated mainly in infiltrating T lymphocytes in lung tissues from patients with IPF.[14] Therefore, we hypothesized that Fas-FasL pathway may be involved in the pathogenesis of IPF.
