  Blue Feulgen Stain
The combination of acid hydrolysis and subsequent aldehyde staining with a Schiff reagent constitutes the Feulgen procedure (Feulgen was one of two individuals who first developed and described the procedure).
Schiff regent is commonly used in histology for both the Feulgen procedure and for the Periodic Acid Schiff (PAS) procedure.
It is also possible to combine ImagePath’s blue Feulgen staining of the cell nucleus with a standard PAS stain for cell glycogen.

The Feulgen method is highly specific for DNA, so the results show that the regions in the nucleus whose density is greatly increased by treatment with uranyl acetate are those which contain substantial amounts of DNA.
To find out whether it is the DNA itself which is stained by uranyl acetate, or whether it is some other component associated with it, we have measured by direct weighing the uptake of stain by samples of purified DNA, of nucleohistone, and of histone, exposed to aqueous solutions of uranyl acetate of various concentrations.
Finally, the Feulgen staining experiment indicates that at least a substantial part of the nucleic acid originally present in several tissues survives the preparative procedures and is available in the sections.

Terms like Feulgen stain and Feulgen reaction is presently the only staining technique which is stoichiometric for DNA, which means that staining of DNA is both quantitative and specific.
The cells are first stained with an alkaline phosphatase technique including a monoclonal antibody specific to a protein in at least one of the cell's cytoplasm or on a cell membrane, thereby marking any cells including the protein as to type.
A second staining of the DNA in the nucleus is accomplished by a Feulgen technique that destroys the cell cytoplasm.

The present study was undertaken to compare the cytoprotective effect of melatonin on the heart with that of recombinant human growth hormone (hGH), a treatment of demonstrable cytoprotective effectivity in experimental cardiac infarction [2,17,18].

Despite of a huge amount of literature on the Feulgen reaction for DNA relatively few investigations deal with the special needs of plant scientists.
In the present study, we re-evaluated recently published Feulgen densitometric data on genome size and its infraspecific variation in Arachis hypogaea, and also conducted measurements in one accession of its wild relative A. monticola.
The methods applied were propidium iodide flow cytometry and Feulgen densitometry using Pisum sativum as an internal standard.

The Blue Feulgen DNA Ploidy Analysis Staining Kit is a histochemical reagent kit for quantifying the nuclear DNA content in cells.
The basis of the Feulgen staining procedure is the use of an acid hydrolysis step to remove purines from the deoxyribose backbone of the DNA molecule.
The newly exposed deoxyribose sugars have an aldehyde group, and this can then be identified by staining with a Schiff reagent (in this case, an aqueous solution of cresyl-violet and sulfurous acid used to test for the presence of aldehydes).

Although German physician Robert J. Feulgen published seventy research papers about the chemistry of physiology, histochemistry, and wrote a review of nucleic acid chemistry titled Chemie und Physiologie der Nucleinstoffe (Chemistry and Physiology of Nucleic Acids), he is best known for his discovery of a method for staining nucleic acid, now termed a Feulgen reaction.
Feulgen was born in Werden, Germany, into a working class family, Feulgen paved his way to the medical school through an outstanding student performance that opened to him the doors of the University of Freiburg at Breisgau in 1905.
His medical residency was taken in the City Hospital of Kiel, where he wrote his dissertation on the purine metabolism of patients with chronic gout.
