Protein folding - Wikipedia, the free encyclopedia
While these macromolecules may be seen as "folding themselves," in fact their folding depends a great deal on the characteristics of their surrounding solution, including the identity of the primary solvent (either water or lipid inside cells), the concentration of salts, the temperature, and molecular chaperones.
During the folding process, the number of hydrogen bonds does not change appreciably, because for every internal hydrogen bond in the protein, a hydrogen bond of the unfolded protein with the aqueous medium has to be broken.
Some proteins never fold in cells at all except with the assistance of chaperone molecules, that isolate individual proteins so that their folding is not interrupted by interactions with other proteins.

The folding has also fractured the limestone, and the lines of weakness thus created were later exploited by marine erosion, forming small arches in the limestone.
Over time, folding can assume highly complicated forms, as can sometimes be seen in the rock layers of cliff faces or deep cuttings in the rock.

A comparison between the observed folding mechanism and that of a peptide b-hairpin analog suggests that although native topology defines the character of the folding landscape, the statistical weighting of potential folding pathways is determined by the chemical nature of the polymer.
From uncoupled ensemble dynamics simulations in unperturbed folding conditions, we report on 1), competing pathways between the folded and unfolded regions of the conformational space; 2), observed non-native stacking and basepairing traps; and 3), a helix unwinding-rewinding mode that is differentiated from the unfolding and folding dynamics.
The folding mechanism of villin is most consistent with the hydrophobic collapse view of folding: the molecule collapses non-specifically very quickly (20 ns), which greatly reduces the size of the conformational space that needs to be explored in search of the native state.

 BC Online: 2D - Protein Folding/Stability
Folding occurs in the ms to second range, which limits the ability to study the presence of intermediates in the process.
An understanding of protein folding can not be separated from an understanding of protein stability, and an understanding of the nature of the native and denatured state.
In studying protein folding and stability/structure of the native and denatured states, both equilibrium (thermodynamic) and timed (kinetic) measurements are made.

 Folding and Unfolding (Erik Demaine)
Folding silhouettes and wrapping polyhedra: We show that "origami design is always possible," in the sense that if you desire a given polyhedron (or a flat silhouette) you can fold it out of a sufficiently large piece of paper.
The basic constraints in folding a piece of paper are that the paper is folded continuously (no ripping), while preserving distances along its surface (no stretching), and not causing the paper to self-intersect (no crossing).
In fact some of my early work in "folding" was on rock folding or the study of tectonic plates, in particular plate subduction, at the Geodynamics Group at Dalhousie University.
