GABA receptor - Wikipedia, the free encyclopedia
GABA is the chief inhibitory neurotransmitter in the mammalian brain.
receptors were initially distinguished by their sensitivity to the ligand bicuculline with the former being antagonized by it while the latter were insensitive.
The GABA A receptor is a member of the Cys-loop ligand-gated ion channel superfamily which also includes the glycine, 5-hydroxytryptamine (5-HT, serotonin), and nicotinic acetylcholine receptors.

GABA A receptor - Wikipedia, the free encyclopedia
The receptor is a multimeric transmembrane receptor that consists of five subunits arranged around a central pore.
The ligand GABA is the endogenous compound that causes this receptor to open; once bound to GABA, the protein receptor changes conformation within the membrane, opening the pore in order to allow chloride ions (Cl-) to pass down an electrochemical gradient.

The GABA-a receptor is similar to acetylcholine receptor in that it is related to an ion channel.
Binding of GABA to this receptor increases the permeability to chloride ion which causes a hyperpolarization of the neuron or inhibition.
The feedback inhibition from the GABA neurons of the globus pallidus and putamen to the dopaminergic neurons of the substantia nigra is an important modulating force on the activity of the dopamine neurons.

GABA is produced by decarboxylation of glutamate mediated by the enzyme glutamic acid decarboxylase (GAD).
GABA has 2 types of receptors, GABA-A and GABA-B. When GABA-A receptor is stimulated, chloride channels open to allow the influx of negative ions (ie, chloride) into the neuron and cause hyperpolarization, moving the membrane potential further from the cell-firing threshold.
GABA is metabolized by transamination in the extracellular compartment by GABA-transaminase (GABA-T).

Once the neurotransmitter molecule reaches the target cell, it binds to its receptor, and its influence on that cell is mediated by the specific type of receptor found on the cell surface.
GABAA receptor is a complex assembly of proteins, made up of five protein subunits with a hole in middle-like a tube made of Legos-to form the chloride channel.
The receptor spans the membrane of the nerve cell, so that the channel extends from outside to inside the cell and chloride can move in if the channel is open.
