GFAP definition - Medical Dictionary definitions of popular medical terms
The degenerative brain condition called Alexander disease is caused by mutation in GFAP (glial fibrillary acidic protein).
The altered GFAP protein may block the normal assembly of intermediate filaments and other proteins that contribute to the structure of astrocytes.
The gene for GFAP is on chromosome 17 in band 17q21.

NIOSH Update / Further Evidence of GFAP as a Useful Marker is Offered in New Study by NIOSH, Partner Researchers
NIOSH researchers and their partners have been investigating the question of whether various occupational exposures are associated with a risk of neurological disease, using a study population of Japanese-American men who were born between 1900 and 1919.
In this study, levels of GFAP, a sensitive marker of neural damage, were measured in brain tissue samples of deceased men who had been diagnosed with dementia, and of deceased men who had not been diagnosed with dementia.
Further, the researchers showed that higher GFAP levels were associated with a lower performance on a test of cognitive function.

Vimentin is the intermediate filament protein that is expressed early in astrocytic development, GFAP is expressed as astrocytes mature.
The thrust of recent research has been to identify the specific amino acid residues that are phosphorylated in GFAP and vimentin so that the phosphorylation of each may be studied independently.
To accomplish this goal a double knockout of the GFAP gene is required in these diploid cells.

This GFAP is a 50-kD intracytoplasmic filamentous protein that constitutes a portion of, and is specific for, the cytoskeleton of the astrocyte.
It is known that early in neuroembryogenesis, astrocytes may synthesize vimentin instead of GFAP as their primary IF.[7,109] The switch to GFAP predominance occurs prenatally in human astrocytes and is caused by factors that have yet to be elucidated.
Because GFAP is a cytoskeletal element that appears to play an important role in astrocytic differentiation and because it is the most specific marker for cells of astrocytic lineage, several groups have studied how the modulation of GFAP expression might affect astrocytoma growth and differentiation.

The expression of GFAP in SVZ stem cells has lead investigators to call them astrocytes, a generic term for glia expressing GFAP.
These GFAP-expressing cells express several features in common with astrocytes; these include the presence of glycogen granules, close contacts with blood vessels, ensheathment of neuronal elements (neuroblasts in the SVZ and synapses elsewhere in the brain), and glutamate transporters.
Furthermore, ependymal cells, which are not astrocytes, express GFAP and derive from radial glia but are postmitotic.

Cayman Chemical | GFAP (human) EIA Kit | Description
Glial fibrillary acidic protein (GFAP) is a 40 - 53 kDa monomeric molecule found only in adult glial cells of the central nervous system (CNS) and represents the major part of the cytoskeleton of astrocytes.
Traumatic injury to the adult CNS results in a rapid inflammatory response by the resident astrocytes, characterized mainly by hypertrophy, proliferation (of astrocytes), and increased GFAP expression, resulting in astrogliosis.
Thus, GFAP is considered to be a reliable cell-specific biomarker for monitoring neuronal activity under developmental and pathological conditions, such as brain injury, and retinal stress.
