This tiny amount of GMCSF took ten years to isolate, and one rule of protein chemistry is that if you get two proteins on a gel, the one you're interested in will be the smaller one.
The GMCSF molecule binds to its specific receptor, setting up a signal which regulates the expression of new genes in the target cell.
Experimentally we gave GMCSF and surprisingly we found that the white blood cellcount began to rise; it rose from a level of zero up to many thousands, an as soon as the neutrophil count began to rise, the patient's fever went away and blood pressure returned to normal.

GMCSF: (granulocyte-macrophage colony-stimulating factor) is indicated in bome marrow insufficiency caused by anti neoplasm therapy.
GMCSF elicits these functions by activating a receptor consisting of a ligand-specific -chain and a -chain, which is common for GMCSF, IL-3 and IL-5.
Using a yeast two-hybrid system, we identified the -chain of the GMCSF receptor as putative interaction partner of IB kinase, one of the central signaling kinases activating the transcription factor NF-B. Using endogenous protein levels of endothelial cell extracts, we could verify the interaction by co-immunoprecipitation experiments.

Unlike IL-4, there is a fairly large patch of hydrophobic residues on one side of the protein surface.
It is interesting to note that most of the Prolines in the GMCSF structure are all located on one side of the molecule surface, mostly in the loop regions and thus are exposed to some degree, except one is in one of the helices and buried.
It is clear from the above table that not all the "conserved" Leucines are located in helix because the various helices of the different cytokines have different lengths.
