The town was renamed in honor of General Edmund Pendleton Gaines, a hero of the War of 1812 who held a command in Georgia during the ensuing struggles with the Creeks.
Gainesville prospered as agricultural activity increased along the rich valleys of the Oconee and Chattahoochee rivers, where abundant corn fattened livestock for the citizens' tables and allowed for homemade production of corn whiskey.
In the late 1850s, as the division between the North and the South deepened, most residents of Gainesville and Hall County were opposed to secession.

While the Gainesville–Hall County area has long functioned as an agricultural, commercial, and financial center for northeast Georgia, as late as the 1950s it lacked a public higher educational institution to meet the escalating demands of a community experiencing rapid economic and demographic growth.
Aware of this urgent need, Gainesville civic leaders responded enthusiastically to the recommendations of Governor Carl Sanders's Commission to Improve Education, as well as to encouragement from the University System of Georgia Board of Regents, by launching a campaign to build local support for a community college in Hall County.
Among the key figures in the effort to mobilize community resources were Mathis, Georgia Highway Commissioner Loyd Strickland, businessman Jesse Jewell, and James A. Dunlap, chair of the Board of Regents—all of them citizens of the Gainesville area.
