In a preferred embodiment of the invention, the composition comprises (a) pectin or a derivative of pectin or a mixture thereof and (b) galactomannan.
When pectin or galactomannan (such as guar gum or locust bean gum) is used alone as a drug carrier, e.g., coating material, the coating is easily dissolved and/or disintegrated in simulated gastric fluid (SGF) and simulated intestinal fluid (SIF).
Because both the pectin and galactomannan components of the composition of the present invention are specifically degraded in the colon, there is a burst of drug release in the colon due to the synergic effect of the colonic enzymatic activities.

Galactomannans obtained from guar (Cyamopsis tetragonoloba), lucern (Medicago sativa) and fenugreek (Trigonella foenum-graecum) containing high proportions of galactose are mentioned as examples which can be used as starting materials in the process described in EP-A- 0 121 960.
They yield galactomannans which have improved properties and can be used, for example as substitutes for locust bean (carob) gum, which has more favourable properties, such as gelling properties, in particular with other polysaccharides, than untreated guar gum has.

The fenugreek galactomannan (hydrocolloid) can be used as a stabilizer and/or additive in a variety of foods, while with further development, the saponin fraction could also be employed as an effective emulsifier in cosmetics.
Recently, physiological experiments carried out in animals and humans have shown that both galactomannan and saponin are effective in lowering cholesterol and glucose levels in the blood.
The galactomannan has very good emulsification capabilities compared to other water soluble hydrocolloids and is a water soluble gum, having surprisingly unexpected surface activity.

A model of primary pulmonary aspergillosis in rabbits was developed to reproduce the persistent levels of profound granulocytopenia and the histopathologic features of bronchopneumonia, vascular invasion, and hemorrhagic infarction encountered in humans.
D-mannitol was detectable in bronchoalveolar lavage fluid by gas-liquid chromatography/mass spectroscopy, and galactomannan was measurable in serum by latex agglutination immunoassay.
Thus, D-mannitol in lavage fluid and galactomannan in serum may be useful markers of pulmonary aspergillosis, and liposomal amphotericin B was significantly more effective and safer than desoxycholate amphotericin B for treatment of pulmonary aspergillosis in profoundly granulocytopenic rabbits.
