The Knudson hypothesis is the hypothesis that cancer is the result of accumulated mutations to a cell's DNA.
It was first formulated by Alfred G. Knudson in 1971, and led indirectly to the identification of cancer-related genes.
Knudson performed a statistical analysis on cases of retinoblastoma, a tumour of the retina which occurred both on children as a hereditary disease and in adults in a sporadic fashion.

Later, Professor Knudson focused his hypothesis on the RB gene itself as well as other genes involved in different cancer syndromes like Wilm's tumor and renal cell carcinomas using the Eker rat, a perfect model for hereditary renal cell carcinoma, developed by Reidar Eker at the Norwegian Radium Hospital.
Professor Knudson’s theory was that each hit must arise in one of the two copies of the gene involved, suggesting that both copies of the gene must be inactivated for a tumor to form.

George Archer is survived by his wife of 44 years, the former Donna Garman, who was also his swing coach; two daughters, Elizabeth Klein of Tacoma, Wash., and Marilyn DeChambrier of Gilroy; seven grandchildren; two sisters, Ruth Callas of Healdsburg, Calif., and Dorothy Scott of San Francisco; and a brother, Tom of Guerneville, Calif.
